Summary: This study revealed the role of APOBEC3B in prostate cancer (PCa) chemoresistance and its dysregulated expression mechanism. Results showed that APOBEC3B was upregulated in docetaxel-resistant PCa cells and its knockdown inhibited cell proliferation and docetaxel resistance. Bioinformatics and luciferase report analysis confirmed that miR-138-5p targeted APOBEC3B. Overexpression of miR-138-5p suppressed cell proliferation and docetaxel resistance, while inhibition of miR-138-5p reversed this process. Furthermore, upregulation of APOBEC3B expression desensitized PCa cells to docetaxel treatment in docetaxel-resistant cells overexpressing miR-138-5p. In summary, miR-138-5p regulates PCa cell proliferation and chemoresistance by targeting the 3'-UTR of APOBEC3B, providing new insights and therapeutic targets for PCa treatment.

Summary: In our study, we discovered that extracellular vesicles (EVs) play a crucial role in aging, as they can induce chronic inflammation by activating immune cells. We found that EVs derived from senescent cells contain miR-30b-5p, which downregulates SIRT1 and enhances pro-inflammatory cytokine production. Additionally, we observed increased levels of miR-30b-5p in both the aging animal model and EVs harvested from tissues. EVs carrying pro-inflammatory signals can alter immune cell activity in the aging microenvironment, leading to an increase in inflammation.
