Oxidative metabolism of BDE-47, BDE-99, and HBCDs by cat liver microsomes: Implications of cats as sentinel species to monitor human exposure to environmental pollutants

The in vitro oxidative metabolism of 2,2',4,4'-tetrabromodiphenyl ether (BDE-47), 2,2',4,4',5-pentabromodiphenyl ether (BDE-99), and individual alpha-, beta- and gamma-hexabromocyclododecane (HBCD) isomers catalyzed by cytochrome P450 (CYP) enzymes was screened using cat liver microsomes (CLMs). Six hydroxylated metabolites, namely 4-hydroxy-2,2',3,4'-tetrabromodiphenyl ether (4-OH-BDE-42), 3-hydroxy-2,2',4,4'-tetrabromodiphenyl ether (3-OH-BDE-47), 5-hydroxy-2,2',4,4'-tetrabromodiphenyl ether (5-OH-BDE-47), 6-hydroxy-2,2',4,4'-tetrabromodiphenyl ether (6-OH-BDE-47), 4'-hydroxy-2,2',4,5'- tetrabromodiphenyl ether (4'-OH-BDE-49), and 2'-hydroxy-2,3',4,4'-tetrabromodiphenyl ether (2'-OH-BDE-66), were identified and quantified after incubation of BDE-47. A di-OH-tetra-BDE was also found as metabolite of BDE-47 with CLMs. 5-OH-BDE-47 was the major metabolite formed. Five hydroxylated metabolites (3'-hydroxy-2,2',4,4',5-pentabromodiphenyl ether (3'-OH-BDE-99), 5'-hydroxy-2,2',4,4',5-pentabromodiphenyl ether (5'-OH-BDE-99), 6-hydroxy-2,2',4,4',5-pentabromodiphenyl ether (6-OH-BDE-99), 6'-hydroxy-2,2',4,4',5-pentabromodiphenyl ether (6'-OH-BDE-99), and 4'-hydroxy-2,2',4,5,5'-pentabromodiphenyl ether (4'-OH-BDE-101) were formed from BDE-99 incubated with CLMs. Concentrations of BDE-99 metabolites were lower than those of BDE-47. Four or more monohydroxylated HBCD (OH-HBCDs), four or more di-hydroxylated HBCD (di-OH-HBCDs), five or more mono-hydroxylated pentabromocyclododecanes (OH-PBCDs), and five or more di-hydroxylated pentabromocyclododecenes (di-OH-PBCDs) were detected after incubation of alpha-, beta-, or gamma-HBCD with CLMs. No diastereoisomeric or enantiomeric enzymatic isomerisation was observed incubating alpha-, beta- or gamma-HBCD with CLMs. Collectively, our data suggest that (i) BDE-47 is metabolized at a faster rate than BDE-99 by CLMs, (ii) OH-HBCDs are the major hydroxylated metabolites of alpha-, beta- and gamma-HBCD produced by CLMs, and (iii) the oxidative metabolism of BDE-47 and BDE-99 is different by cat and human liver microsomes. This suggests that cats are not a suitable sentinel to represent internal exposure of PBDEs for humans, but is likely a promising sentinel for internal HBCDs exposure for humans. Transfer of Microplastics in Terrestrial and Aquatic Food Webs: The Impact of E-Waste Debris and Ecological Traits

Summary: This study determined the abundances of microplastics (MPs) in various terrestrial and aquatic species from an abandoned e-waste recycling site. The abundances of MPs in wildlife were found to be positively correlated with body weight per individual but negatively correlated with body weight per body weight. The composition of polymer types in terrestrial vertebrates, especially birds, was more complex compared to other organisms.

A Production and Fractionation Protocol for Polyvinyl Chloride Microplastics

Summary: Concerns about microplastics in the environment have grown, leading scientists to conduct thorough studies. A cryogenic grinding and wet-sieving method was developed to efficiently produce and fractionate microplastics. With this method, polyvinyl chloride (PVC) microplastics could be produced with specific particle sizes and easily separated into different size ranges. The method has the potential to be applied to other types of plastics as well.

Metabolic signature of HepaRG cells exposed to ethanol and tumor necrosis factor alpha to study alcoholic steatohepatitis by LC-MS-based untargeted metabolomics

Summary: Despite being common, alcoholic liver disease remains poorly understood, especially in its early stages. In this study, alcoholic steatohepatitis was simulated in vitro to investigate the metabolic alterations and identify affected pathways. The researchers discovered key findings, such as the influence of TNF-alpha on hepatic triglycerides and the detection of FAHFA-containing triglycerides in human hepatocytes, which may play an important role in the progression of alcoholic steatohepatitis. Additionally, ethoxylated phosphorylcholine was identified as a potential new biomarker of ethanol exposure.

Assessment of silicone wristbands for monitoring personal exposure to chlorinated paraffins (C8-36): A pilot study

Investigating the Potential of Drift Tube Ion Mobility for the Analysis of Oxidized Lipids

Summary: Epilipids, a subset of the lipidome, play a crucial role in regulating redox lipid metabolism and influencing cell fate decisions. Ion mobility spectrometry (IMS) combined with liquid chromatography-high-resolution mass spectrometry (LC-HRMS) can improve the peak capacity and separation ability for the analysis of oxidized complex lipids. This study demonstrates the advantages and limitations of implementing IMS in LC-HRMS workflows and provides insights into the annotation of oxidized complex lipids.

Identification and characterization of quaternary ammonium compounds in Flemish indoor dust by ion-mobility high-resolution mass spectrometry

Summary: Quaternary ammonium compounds (QACs) in European indoor dust were identified, characterized, and semi-quantified using ion-mobility high-resolution mass spectrometry. Targeted QACs (21) and suspect QACs (17) were detected with high detection frequencies. The concentrations of individual QACs were estimated and daily intakes for adults and toddlers were calculated. This study provides important insights into the potential human exposure to QACs.

An international investigation of chlorinated paraffin concentrations and homologue distributions in indoor dust*

Summary: In this study, dust samples from Japan, Australia, Colombia, and Thailand were analyzed for different chain lengths of chlorinated paraffins. The results showed that chlorinated paraffins were detected in all samples, with the highest concentrations found in samples from Colombia and Thailand. Based on current toxicological data, it was determined that short-chain and medium-chain chlorinated paraffins in indoor dust do not pose a health risk through ingestion.

A retrospective investigation of feather corticosterone in a highly contaminated white-tailed eagle (Haliaeetus albicilla) population

Summary: English Summary: Exposure to persistent organic pollutants (POPs), such as organochlorines (OCs) and polybrominated diphenyl ethers (PBDEs), can have adverse health effects on wildlife. White-tailed eagles in the Baltic ecosystem have suffered population declines due to severe exposure to DDT and PCB. In this study, feathers from breeding WTE pairs were analyzed to investigate the annual variations in feather corticosterone (fCORT), POPs, and dietary proxies. The results did not support fCORT as a relevant biomarker of contaminant-mediated effects in WTEs, but fCORT can be used as a non-destructive assessment of long-term stress physiology in wild raptors.

Hair as an alternative matrix to assess exposure of premature neonates to phthalate and alternative plasticizers in the neonatal intensive care unit

Summary: This study reports metabolites of phthalate and alternative plasticizers in neonatal hair samples as biomarkers for exposure to these plasticizers. Results showed that hair sampled from premature neonates after a NICU stay contained significantly higher metabolite concentrations of both phthalates and alternative plasticizers compared to healthy control neonates. Prolonged NICU exposure to respiratory support devices and gastric tubes was correlated with increased concentrations in hair samples.

The model accurately characterized the structure of the wetland food web and improved the reliability of the trophic magnification factor. The study revealed three interlocking food webs in the wetland and found that the kingfisher food web had the highest biomagnification capacity.
