Summary: COVID-19 poses a serious threat to public health, and a wearable iontophoresis-driven microneedle (MN) patch is developed for active and efficient transdermal vaccine delivery. The patch consists of polyacrylamide/chitosan hydrogels with excellent properties, and the delivery efficiency is greatly improved by the synergistic effect of MN puncture and iontophoresis. In vitro and in vivo studies demonstrate controlled delivery of ovalbumin and a stronger immune response compared to traditional intramuscular injection, with minimal biosafety concerns. This low-cost and user-friendly delivery system has great potential for vaccine self-administration at home.

Summary: Oral inflammatory disease (OID), particularly oral ulcers, is a common oral lesion that affects quality of life and can lead to oral cancer. Traditional treatment methods for oral ulcers, such as localized hormone injections, can cause pain and fear. To address this, a soluble hyaluronic acid (HA) microneedle patch containing betamethasone 21-phosphate sodium (BSP) and betamethasone 17,21-dipropionate (BDP) was developed. The patch showed mechanical strength to penetrate the mucosa and released BSP and BDP into the ulcer base rapidly. It exhibited wound healing-promoting and anti-inflammatory effects, providing a new prospect for oral ulcer treatment.

The use of HA microneedles loaded with melittin showed promising results in inhibiting RA progression in animal models, with high mechanical strength and sustained release properties. Furthermore, the microneedles were able to suppress pro-inflammatory cytokines and increase regulatory T cell percentage, indicating potential as a new therapy for RA and other autoimmune diseases.
