Aphloia theiformis is traditionally used in Mauritius, Madagascar, and Reunion Island for treating several diseases. In this study, various extraction solvents and schemes were applied for the recovery of antioxidant rich fractions from the leaves of A. theiformis. The products were evaluated for their antioxidant capacity using well known in vitro assays. Major compounds were characterized by UPLC-QTOF-MS. Hydrophilic extracts of A. theiformis demonstrated strong antioxidant properties, which are comparable with the synthetic antioxidant Trolox. UPLC analysis confirmed mangiferin as the main secondary metabolite of A. theiformis. Tormentic and hydroxytormentic acids as well as their isomers were also abundant in A. theiformis extracts and fractions, while their amounts were determined for the first time. The most potential extract was further separated into the fractions by liquid-liquid extraction and by precipitation at low temperature. Antioxidant capacity and composition of secondary metabolites of derived fractions were determined. Some of the fractions possessed remarkable antioxidant capacity, comparable to pure mangiferin. The results obtained reveal high potential of A. theiformis for recovery of natural antioxidants and other bioactive phytochemicals, particularly mangiferin.

Supercritical CO2 Extraction of Narcissus poeticus L. Flowers for the Isolation of Volatile Fragrance Compounds

Renata Baranauskiene, Petras Rimantas Venskutonis

Summary: The study developed a method for extracting fragrance substances from Narcissus poeticus using pure supercritical CO2 and a mixture of CO2 with 5% ethanol. The results showed that the extracts contained various volatile compounds, including benzyl benzoate, alpha-terpineol, and limonene. The findings are significant for the further extraction of fragrance substances from Narcissus poeticus using supercritical CO2.

Cranberry Pomace Extract Exerts Antiviral Activity against Zika and Dengue Virus at Safe Doses for Adult Zebrafish

Laura Tamkute, Juliano G. Haddad, Nicolas Diotel, Philippe Despres, Petras Rimantas Venskutonis, Chaker El Kalamouni

Summary: In this study, it was found that Cranberry Pomace extract can block the infection of dengue virus and zika virus in human cells, showing good antiviral effects at non-cytotoxic concentrations. By directly acting on viral particles, the extract can prevent the attachment of viruses to the cell surface. Furthermore, it was also demonstrated to be safe and tolerable in a zebrafish model.

Summary: In this study, synthetic peptide-based ELISA assays were used to identify linear B-cell epitopes in the spike protein that contribute to antibody response in individuals vaccinated with the Pfizer-BioNTech COVID-19 vaccine. The synthetic peptides S2P6 and S1P4 were recognized by the immune sera from vaccine recipients, suggesting their potential usefulness in measuring antibody response to COVID-19 mRNA vaccines. The S2P6 peptide has been shown to be immunogenic in mice, raising questions about its role in the protective efficacy of the vaccine.

Health Beneficial Phytochemicals in Dioscorea caucasica Lipsky Leaves and Tubers and Their Inhibitory Effects on Physiologically Important Enzymes

Ausra Adomeniene, Audrius Pukalskas, Ona Ragazinskiene, Petras Rimantas Venskutonis

Summary: This study identified 41 compounds in the leaf and tuber hydroethanolic extracts of Dioscorea caucasica using UPLC-QTOF/MS. The main phytochemicals in the leaves were hydroxycinnamates and flavonoids, while steroidal glycosides, fatty acids, and carbohydrates were found in the tubers. The leaf extracts showed strong inhibitory activity against key enzymes, including alpha-amylase, alpha-glucosidase, acetylcholinesterase, and angiotensin-converting enzyme. These findings contribute to the development of new functional foods and nutraceuticals.

Stability of Dengue 2 Nonstructural Glycoprotein 1 (NS1) Is Affected by the Nature of Basic Residue at Position NS1-324

Eva Ogire, Chaker El-Kalamouni, Philippe Despres, Marjolaine Roche

Summary: Dengue is a common viral disease transmitted by mosquitoes, and the NS1 protein of DENV-2 plays a crucial role in virus replication and infection. In this study, the impact of mutations at positions 272 and 324 on NS1 protein stability and secretion was investigated. It was found that replacing arginine with lysine at position 324 in the DES-14 strain resulted in decreased protein stability and secretion, leading to oxidative stress and pro-inflammatory cytokine activation in Huh7 cells. The ubiquitin-proteasome proteolytic pathway may be involved in the stability of DENV-2 protein containing a lysine residue at position 324.
