Summary: This review focuses on the sources of mitochondrial proteotoxicity and recent cellular strategies to combat such stress and dysfunction of the organelle. The communication between mitochondria and neighboring subcellular compartments during proteotoxic stress, such as the endoplasmic reticulum and cytosol, has been explored. Alterations of crucial inter-organelle connections like ER-mitochondria contact sites and their implications on mitochondrial signaling activity like Ca2+ signaling have been discussed. Furthermore, pathological conditions, particularly neurodegenerative disorders associated with mitochondrial proteotoxicity and Ca2+ dysregulation, have been outlined.

Summary: Cadmium (Cd2+) exposure-induced cytotoxicity and renal injury can be protected against by activating the calcium-sensing receptor (CaSR) to restore the intracellular Ca2+ homeostasis through the sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) channel. SERCA2 degradation caused by Cd2+ can be inhibited by the proteasome inhibitor MG132, indicating the involvement of the proteasomal protein degradation pathway in maintaining SERCA2 stability. Targeting SERCA2 and the associated proteasome may provide a new therapeutic approach for preventing Cd2+-induced cytotoxicity and renal injury.

Summary: This study found that oxidative stress, myeloperoxidase, and endoplasmic reticulum stress are increased in a rat model of bronchopulmonary dysplasia (BPD). Treatment with tunicamycin or hyperoxia further increased endoplasmic reticulum stress and decreased lung complexity, resembling the BPD phenotype. Inhibition of endoplasmic reticulum stress with tauroursodeoxycholic acid improved lung complexity and decreased myeloid cells and myeloperoxidase levels. These findings suggest a causal role of endoplasmic reticulum stress in BPD and highlight the potential of tauroursodeoxycholic acid as a treatment.

Summary: The recently defined type of cell death, ferroptosis, has attracted significant attention as a potential new approach to cancer treatment due to its more immunogenic nature compared with apoptosis. In this study, a compound called Diplacone (DP), found in the fruit of Paulownia tomentosa, was found to have anti-inflammatory and anti-radical activity, and potential anticancer activity against A549 human lung cancer cells. DP induced a form of cytotoxicity distinct from apoptosis, accompanied by extensive mitochondrial-derived cytoplasmic vacuoles.
