Summary: This study revealed that DGUOK-AS1 promotes proliferation of cervical cancer cells by regulating the miR-653-5p/EMSY pathway. miR-653-5p, which is lowly expressed in cervical cancer cells, is sponged by DGUOK-AS1 and inhibits proliferation of cervical cancer. EMSY, a target gene of miR-653-5p, is regulated by DGUOK-AS1 through sponging miR-653-5p and shows significantly high expression in cervical cancer cells.

It promotes cell migration and tumor metastasis, while knockdown of hsa_circ_0003258 has the opposite effect. Mechanistically, hsa_circ_0003258 functions by sponging miR-653-5p and increasing the expression of ARHGAP5. Additionally, hsa_circ_0003258 physically binds to IGF2BP3 and enhances HDAC4 mRNA stability, activating the ERK signaling pathway and triggering EMT programming.

Summary: Our study investigated the role of lncRNA TDRKH-AS1 in breast cancer (BC). We found that TDRKH-AS1 was upregulated in BC tissues and cell lines, and its high expression was associated with advanced stages and poor outcomes in BC patients. Knockdown of TDRKH-AS1 inhibited BC cell proliferation and invasion. Mechanistically, TDRKH-AS1 acted as a sponge for miR-134-5p, thereby promoting the expression of cAMP response element-binding protein 1 (CREB1), a downstream target of miR-134-5p. In vivo experiments confirmed the tumor-promoting role of TDRKH-AS1 in BC.
