1,2-Bis(4-pyridyl)ethane (bpa) and 1,2-bis(4-pyridyl)ethene (bpe) were used for the synthesis of polynuclear silver(I) complexes, {[Ag(bpa)]NO3}(n) (1), {[Ag(bpa)(2)](CF3SO3H2O)-H-.}(n) (2) and {[Ag(bpe)]CF3SO3}(n) (3). In complexes 1-3, the corresponding nitrogen-containing heterocycle acts as a bridging ligand between two Ag(I) ions. In vitro antimicrobial activity of these complexes, along with the ligands used for their synthesis, was evaluated against the broad panel of Gram-positive and Gram-negative bacteria and fungi. The silver(I) complexes 1-3 showed selectivity towards Candida spp. and Gram-negative Escherichia coli in comparison to the other investigated bacterial strains, effectively inhibiting the growth of four different Candida species with minimal inhibitory concentrations (MICs) between 2.5 and 25 mu g/mL and the growth of E. coli, with MIC value being 12.5 mu g/mL. Importantly, complex 2 significantly reduced C. albicans filamentation, an essential process for its pathogenesis. Antiproliferative effect on the normal human lung fibroblast cell line MRC-5 was also evaluated with the aim of determining the therapeutic potential of the complexes 1-3. The interactions of these complexes with calf thymus DNA (ctDNA) and bovine serum albumin (BSA) were studied to evaluate their binding activities towards these biomolecules for possible insights on their mode of action.

Expanding the Scope of Polyoxometalates as Artificial Proteases towards Hydrolysis of Insoluble Proteins

Nada D. Savic, David E. Salazar Marcano, Tatjana N. Parac-Vogt

Summary: The study reports the hydrolysis of fully insoluble protein using polyoxometalate complexes as artificial proteases in surfactant solutions for the first time. The results indicate that the hydrolytic selectivity and activity of the POM catalysts strongly depend on the concentration of surfactant.

Understanding the Role of Surfactants in the Interaction and Hydrolysis of Myoglobin by Zr-MOF-808

Charlotte Simms, Nada D. Savic, Kimmy De Winter, Tatjana N. Parac-Vogt

Summary: This study investigates the nanozymatic potential of MOF-808 for protein hydrolysis in the presence of different surfactants. The activity of MOF-808 can be tuned by using appropriate surfactants. Spectroscopic methods were used to study the interactions between surfactant, MOF, and protein.

Regioselective protein oxidative cleavage enabled by enzyme-like recognition of an inorganic metal oxo cluster ligand

Shorok A. M. Abdelhameed, Francisco de Azambuja, Tamara Vasovic, Nada D. Savic, Tanja Cirkovic Velickovic, Tatjana N. Parac-Vogt

Summary: Oxidative modifications of proteins play a crucial role in biotechnology, and metal-catalyzed oxidation reactions have low selectivity. However, the use of discrete inorganic ligands such as polyoxometalates enables efficient and selective protein oxidative cleavage by directing the reaction to specific sites on the protein surface.

Copper(ii) and silver(i) complexes with dimethyl 6-(pyrazine-2-yl)pyridine-3,4-dicarboxylate (py-2pz): the influence of the metal ion on the antimicrobial potential of the complex

Tina P. Andrejevic, Ivana Aleksic, Jakob Kljun, Marta Pockaj, Matija Zlatar, Sandra Vojnovic, Jasmina Nikodinovic-Runic, Iztok Turel, Milos I. Djuran, Biljana D. Glisic

Summary: Dimethyl 6-(pyrazine-2-yl)pyridine-3,4-dicarboxylate (py-2pz) was used as a ligand to synthesize new copper(II) and silver(I) complexes. The structures of the complexes were determined by single crystal X-ray diffraction analysis, revealing the bidentate coordination mode of py-2pz to the metal ions. Silver(I) complexes showed good antimicrobial activity against bacteria and fungi. The binding affinity of the complexes with DNA and protein was studied to understand their antimicrobial mechanism. The copper(II) complexes exhibited biomimetic catalytic activity.

Self-Assembled Protein-Surfactant Nanoaggregates for Tunable Peptide Bond Hydrolysis by Polyoxometalate Nanoclusters

Nada D. Savic, David E. Salazar Marcano, Siene Swinnen, Angelo Mullaliu, Tatjana N. Parac-Vogt

Summary: The development of artificial proteases that can maintain their activity in the presence of surfactants is crucial for studying proteins, as natural proteases only work under specific conditions. This study focuses on the hydrolysis of horse heart myoglobin using a specific artificial protease and different surfactants. The results show that the hydrolytic efficiency of the artificial protease increases in the presence of surfactants, but the fragmentation pattern varies depending on the surfactant used. A multitechnique approach was used to gain a better understanding of the protease activity. A general model for protein/surfactant/POM ternary nanoassemblies was proposed based on the findings.

Exploring the Reactivity of Polyoxometalates toward Proteins: From Interactions to Mechanistic Insights

David E. Salazar Marcano, Nada D. Savic, Shorok A. M. Abdelhameed, Francisco de Azambuja, Tatjana N. Parac-Vogt

Summary: This Perspective discusses the reactivity of polyoxometalates (POMs), a family of soluble anionic metal-oxo nanoclusters, towards proteins. POMs serve as catalysts in various reactions and have potential therapeutic applications. However, the lack of detailed understanding of the reaction mechanisms, especially with complex biological systems like proteins, hinders further developments. The reactions of POMs with peptides and proteins are highlighted to provide a molecular-level understanding of the reaction mechanism.

Summary: In this study, the performance of a metal-oxo cluster alpha-/beta-K6P2W18O62 (WD-POM) as a computed tomography (CT) contrast agent was compared to the standard contrast agent iohexol. Toxicity evaluation of WD-POM was conducted on Wistar albino rats, revealing the maximum tolerable dose and acute intravenous toxicity. The study provides a crucial step in assessing the side effects of polyoxometalate nanoclusters.

Tina P. P. Andrejevic, Darko P. P. Asanin, Bojana V. V. Pantovic, Nevena L. J. Stevanovic, Violeta R. R. Markovic, Milos I. Djuran, Biljana D. Glisic

Summary: Pseudomonas aeruginosa is a significant bacterium involved in severe infections and bioengineering. Metal complexes have been used in medicine for ancient times. This review article focuses on three natural products derived from P. aeruginosa and their ability to form complexes with different metal ions. Studying the coordination properties of these compounds is important for understanding metal ion metabolism and exploring their potential use in medicine.

Summary: In this study, two isostructural ferrocene complexes were synthesized and evaluated for their antimicrobial and cytotoxic activities. Complex 1 showed moderate activity against Candida strains and higher cytotoxicity compared to complex 2. Both complexes had no effect on the survival rate of C. elegans. Complex 2 inhibited filamentation of C. albicans, while complex 1 was more effective in inhibiting biofilm formation. Additionally, the binding affinity of the complexes with DNA and protein was studied.
