Analysis of Let-7 Family miRNA in Plasma as Potential Predictive Biomarkers of Diagnosis for Papillary Thyroid Cancer

The most common histological type of thyroid cancer is papillary thyroid carcinoma (PTC). Radical resection of the thyroid gland is currently the recommended method of treatment. Almost 75% of thyroidectomies performed just for diagnostic purposes are benign. Thus, the confirmation of innovative and more precise noninvasive biomarkers holds promise for the detection of PTC, which may decrease the number of unnecessary thyroid lobectomies. In this work, using the droplet digital PCR (ddPCR) method, we have analyzed the level of five miRNAs (let-7a, let-7c, let-7d, let-7f, and let-7i) in the plasma of patients with PTC and compared them with those of a healthy control group to investigate whether miRNAs also have value in the management of PTC. Levels of four miRNAs, namely let-7a, let-7c, let-7d, and let-7f, were significantly higher in PTC patients than healthy controls. Thus, the analysis of circulating let-7 can be a useful tool and support the currently used methods for PTC diagnosis. However, our observation requires further research on a larger patient group.

Circulating cell free DNA response to exhaustive exercise in average trained men with type I diabetes mellitus

Summary: The study compared the increase in cf DNA after exercise in men with T1DM and healthy controls, finding that there was no significant difference in exercise-induced increment in cf n-DNA between the two groups. However, in the control group, this variable correlated with run distance, Delta neutrophils, Delta creatinine, and Delta creatine kinase.

Summary: Histones and PAD4 levels increase in response to strenuous exercise, but there is no significant correlation between citrullinated histone and aggregation of cell-free nDNA. Positive correlations have been observed between cf nDNA with aggregation and PAD4, lactate with aggregation, and lactate with citrullinated histone.

Summary: Cell-free DNA is commonly found in healthy individuals but increases after physical activity, in patients with trauma, sepsis, and cancer. Current trends in cfDNA applications from clinical trials include oncology, prenatal diagnostics, and organ transplantation. The diagnostic value and accuracy of cfDNA remain controversial, requiring further evaluation before becoming a standard in clinical practice.

Summary: WHO has declared COVID-19 as a global public health emergency. The elderly, pregnant women, and individuals with associated co-morbidities are the most susceptible population groups to infection. Cell-free DNA appears to be a valuable marker for COVID-19 risk assessment, as well as for measuring the complications and underlying health risk factors associated with the disease.

Summary: Multiple myeloma is a disease characterized by the proliferation of malignant monoclonal plasma cells in the bone marrow. Cytokines, such as IL-13 and IL-4, play an important role in the growth and survival of multiple myeloma. This study found that pretreatment levels of IL-13 and IL-4 are predictors of better progression-free survival and overall survival, while IL-1Ra levels negatively impact overall survival in patients treated with bortezomib-based chemotherapy.

Summary: Strenuous exercise can temporarily increase spontaneous oxidant production by circulating phagocytes, while decreasing their response to fMLP. This suggests that exercise can activate mechanisms leading to increased antioxidant activity in regularly training individuals.

Changes in the cardiovascular system in children with pediatric multisystem inflammatory syndrome temporally associated with COVID-19-A single center experience

Summary: Pediatric multisystem inflammatory syndrome temporally associated with COVID-19 (PIMS-TS) is a serious complication in pediatric patients. This study retrospectively analyzed demographic data, clinical course, laboratory tests, and cardiovascular system assessment of 51 patients with PIMS-TS. Results showed that changes in the cardiovascular system in PIMS-TS are diverse, and most patients require close cardiac supervision and long-term follow-up.

Effect of Fatty Acid Amide Hydrolase Inhibitor URB597 on Orofacial Pain Perception in Rats

Summary: This study assessed the effects of fatty acid amide hydrolase inhibitor URB597 in an animal model of orofacial pain caused by tooth pulp stimulation. The results showed that URB597, by inhibiting the metabolism of endocannabinoids and reducing gene expression levels, effectively prevented orofacial pain, providing a promising therapeutic target for its treatment.

Case report: Neonatal diabetes mellitus with congenital hypothyroidism as a result of biallelic heterozygous mutations in GLIS3 gene

Summary: Neonatal diabetes mellitus with congenital hypothyroidism (NDH) syndrome is a rare disease caused by mutations in the GLIS3 gene. This study reports a new case of NDH syndrome and identifies two novel mutations in the GLIS3 gene. The findings provide important insights into the clinical phenotype, treatment approaches, and outcome of infants with GLIS3 mutations.

Effect of Exercise Repetitions on Arylesterase Activity of PON1 in Plasma of Average-Trained Men-The Dissociation between Activity and Concentration

Summary: Exercise can increase plasma antioxidant capacity by stimulating antioxidant enzymes. This study measured the effect of acute exercise on ARE activity of the PON1 enzyme and found that ARE activity remained stable during exercise but decreased after exercise when standardized for PON1 concentration. The ratio of ARE/PON1c returned to baseline levels during rest after each exercise session. Individuals with lower pre-exercise ARE activity may experience a higher inflammatory response to vigorous exercise.

Summary: The endocannabinoid system plays a role in pain processing, and this study aimed to investigate its effects in the orofacial area. The results showed that endocannabinoid AEA can modulate neural structures and may have different regulatory mechanisms than 2-AG in orofacial pain.
