Summary: The mqMSP assay developed in this study showed high sensitivity and specificity in plasma samples from CRC patients, outperforming other methods in detecting early-stage and premalignant diseases, and predicting cancer recurrence. Additionally, the detection of ctDNA in preoperative plasma samples using this method can predict the likelihood of patient recurrence.

Summary: The study explored the pathological role of a nonhistone methyltransferase in gastric cancer (GC), identified EEF1AKMT3 as a novel tumor suppressive methyltransferase, and revealed its mechanism of action through regulating the methylation of MAP2K7 and the degradation of TP53. The findings suggest potential targeted therapy in GC by manipulating the dysregulated EEF1AKMT3/MAP2K7/TP53 signaling axis.

Summary: The study shows that epigenetic therapies can enhance immune response and recruit immune cells to fight ovarian carcinoma by targeting the epigenome, with a role for p53 in this process. Different types of epigenetic therapy result in upregulation of different repetitive elements, and cell lines with TP53 mutations exhibit weaker response.

Summary: This study investigates the role of DNA methylation in tumor immunity and its impact on tumor microenvironment (TME) formation and immune activation. The researchers identified three DNA methylation modification patterns with distinct prognosis and biological behaviors, which correlated with three known phenotypes of immune-inflamed, immune-excluded, and immune-desert. They also demonstrated that a DNA methylation score (DMS) could be used as a predictive tool for immunotherapy. The findings provide valuable insights into TME immune infiltration characteristics and guide more effective and personalized immunotherapy strategies.

Summary: Early cancer detection using cell-free DNA faces challenges of low tumor DNA fraction, molecular heterogeneity, and limited sample sizes. In this study, the authors developed a cost-effective method called cfMethyl-Seq to profile the methylome of cell-free DNA and detect and locate cancer. The approach achieved high sensitivity and accuracy for detecting and identifying the tissue-of-origin of various cancer types.

Summary: DNA methylation at the promoter CpG islands of CHK gene is increased in colon cancer cells and human colon cancer tissues compared to normal healthy counterparts, leading to decreased expression of CHK mRNA. Enhanced levels of DNMT in colon cancer cells and tissues contribute to this process, resulting in increased oncogenic properties in colon cancer cells.

Summary: This study performed a DNA methylome-wide analysis of normal colon organoids derived from familial adenomatous polyposis (FAP) patients compared to healthy subjects. The results showed extensive epigenetic differences in normal colon organoids from FAP patients that were similar to those observed in colorectal cancer (CRC) tumors. The study identified differentially methylated regions (DMRs) and candidate target genes that may be important in CRC tumor development in FAP.

Summary: CXCL14 is a tumor suppressor gene in colorectal carcinoma that is initially activated and then silenced during tumor occurrence and deterioration. Its silencing is primarily caused by promoter hypermethylation. The methylation level of CXCL14 is associated with the anatomical site of tumor occurrence and is correlated with patient age and prognosis. Reversing the hypermethylation of CXCL14 may serve as an epigenetic therapy for colon cancer.

Summary: This study aimed to analyze the endoscopic and pathological characteristics of colorectal laterally spreading tumors (LSTs) to assist in malignant risk stratification. Findings revealed that LST malignancy was correlated with lesion diameter, location, and morphological appearance. Endoscopic submucosal dissection is the preferred treatment for LSTs with a diameter >= 20 mm and cancerization mainly in the rectum with a morphology of LST-G-M.

Summary: The p53 protein acts as a transcription factor that inhibits tumor development, but mutations in the DNA binding domain of the TP53 gene can contribute to tumor formation. These mutations result in a wide range of transcriptional capabilities of the mutant p53 proteins, impacting cancer formation across various tissue types. Inherited and spontaneous TP53 mutations interact with genetic and epigenetic factors to produce diverse phenotypes in patients, which poses challenges for predicting patient outcomes and exploring new therapeutic approaches.

Summary: The authors developed a simple scoring system to differentiate between low-grade and high-grade adenoma/adenocarcinoma in superficial non-ampullary duodenal epithelial tumors. The scoring system, based on multiple factors, achieved a high diagnostic accuracy of 92%, with a sensitivity of 95% and specificity of 93%. There was no significant difference in diagnostic accuracy between novice and expert endoscopists when using the system.

Summary: This study retrospectively reviewed ESD for lesions at NTT Medical Center Tokyo between 2014 and 2019, classifying lesions based on their proximity or infiltration to a diverticulum, and analyzing the outcomes of different ESD strategies. The findings suggest that ESD for colorectal neoplasms in proximity to or extending into a diverticulum is challenging but can be a safe and effective therapeutic option.
