Clinical prognostic value of DNA methylation in hepatoblastoma: Four novel tumor suppressor candidates

Hepatoblastoma (HB) is very rare but the most common malignant neoplasm of the liver occurring in children. Despite improvements in therapy, outcomes for patients with advanced HB that is refractory to standard preoperative chemotherapy remain unsatisfactory. To improve the survival rate among this group, identification of novel prognostic markers and therapeutic targets is needed. We have previously reported that altered DNA methylation patterns are of biological and clinical importance in HB. In the present study, using genome-wide methylation analysis and bisulfite pyrosequencing with specimens from HB tumors, we detected nine methylated genes. We then focused on four of those genes, GPR180, MST1R, OCIAD2, and PARP6, because they likely encode tumor suppressors and their increase of methylation was associated with a poor prognosis. The methylation status of the four genes was also associated with age at diagnosis, and significant association with the presence of metastatic tumors was seen in three of the four genes. Multivariate analysis revealed that the presence of metastatic tumors and increase of methylation of GPR180 were independent prognostic factors affecting event-free survival. These findings indicate that the four novel tumor suppressor candidates are potentially useful molecular markers predictive of a poor outcome in HB patients, which may serve as the basis for improved therapeutic strategies when clinical trials are carried out.

Risk factors for recurrent gastroesophageal reflux disease after Thal fundoplication

Summary: This study found that the postoperative GERD recurrence rate after TF was 5.8%. Univariate analysis showed that the presence of convulsive seizures and the absence of a tracheostomy at the time of TF were significantly associated with recurrence. Multivariate analysis revealed that the presence of convulsive seizures at the time of TF was the only factor significantly associated with recurrence.

Damage control surgery for grade IV blunt hepatic injury with multiple organ damage in a child: a case report

Summary: In this case report, an 8-year-old boy with grade IV blunt hepatic injury was successfully treated using damage control surgery and the ABTHERA Open Abdomen Negative Pressure Therapy System. The boy recovered without neurological sequelae and was discharged after 102 days in the hospital. The use of a damage control strategy in pediatric patients with severe blunt hepatic injury is effective and can help improve outcomes.

Oncogenic Role of ADAM32 in Hepatoblastoma: A Potential Molecular Target for Therapy

Summary: Hepatoblastoma (HBL) is a rare liver malignancy that occurs mainly in early childhood. Although treatment regimens have improved the prognosis, there are still refractory cases and the current treatment often leads to severe side effects. This study demonstrates that ADAM32, a protein involved in various cancer types, is highly expressed in HBL and is associated with poor prognosis. ADAM32 plays a role in cancer cell proliferation, stemness, migration, invasion, and chemotherapy resistance. This suggests that ADAM32 could be a promising target for therapy and provides insights into the molecular mechanisms of HBL carcinogenesis.

In Vitro Transfection of Up-Regulated Genes Identified in Favorable-Outcome Neuroblastoma into Cell Lines

Summary: In this study, we investigated the association between the expression of DHRS3, NROB1, and CYP26A1 genes and the suppression of neuroblastoma cell growth. The overexpression of these genes led to a reduction in cell growth and induced morphological senescence, mainly through pathways related to cellular senescence and cell adhesion. DHRS3 and NROB1 also arrested the cell cycle and induced differentiation and senescence. These findings suggest that the induction of these genes could be a potential therapeutic strategy for suppressing high-risk neuroblastoma growth.

Summary: Hepatoblastoma is the most common malignant liver tumor in children, and small cell undifferentiated histology and low alpha-fetoprotein levels have been associated with poor prognosis. It is important to differentiate rhabdoid tumors from hepatoblastoma with small cell undifferentiated component or low alpha-fetoprotein levels, as the treatment differs significantly. When rhabdoid tumors are correctly diagnosed, neither the presence of small cell undifferentiated component nor low alpha-fetoprotein levels are associated with poor prognosis.

COOBoostR: An Extreme Gradient Boosting-Based Tool for Robust Tissue or Cell-of-Origin Prediction of Tumors

COOBoostR uses regional somatic mutation density information and chromatin mark features in an extreme gradient boosting-based machine-learning algorithm. By ranking chromatin marks from different tissue and cell types, COOBoostR can determine the most likely tissue or cell of origin based on the somatic mutation density landscape of a sample.
