Regulation of AMH, AMHR-II, and BMPs (2,6) Genes of Bovine Granulosa Cells Treated with Exogenous FSH and Their Association with Protein Hormones

Anti-Mullerian hormone (AMH) is an important reproductive marker of ovarian reserve produced by granulosa cells (GCs) of pre-antral and early-antral ovarian follicles in several species, including cattle. This hormone plays a vital role during the recruitment of primordial follicles and follicle stimulating hormone (FSH)-dependent follicular growth. However, the regulatory mechanism of AMH expression in follicles is still unclear. In this study, we compared the expression of AMH, AMHR-II, BMP2, BMP6, FSHR, and LHCGR genes during follicular development. In-vitro expression study was performed with and without FSH for AMH, AMHR-II, BMP2, and BMP6 genes in bovine GCs which were isolated from 3-8 mm follicles. Association among the mRNA expression and hormone level was estimated. GCs were collected from small (3-8 mm), medium (9-12 mm) and large size (13 to 24 mm) follicles before, during onset, and after deviation, respectively. Further, mRNA expression, hormones (AMH, FSH, and LH), apoptosis of GCs, and cell viability were detected by qRT-PCR, ELISA, flow cytometry, and spectrophotometry. AMH, AMHR-II, BMP2, and FSHR genes were highly expressed in small and medium follicles as compared to large ones. In addition, the highest level of AMH protein (84.14 +/- 5.41 ng/mL) was found in medium-size follicles. Lower doses of FSH increased the viability of bovine GCs while higher doses repressed them. In-vitro cultured GCs treated with FSH significantly increased the AMH, AMHR-II, and BMP2 expression levels at lower doses, while expression levels decreased at higher doses. We found an optimum level of FSH (25 ng/mL) which can significantly enhance AMH and BMP2 abundance (p < 0.05). In summary, AMH, AMHR-II, and BMP2 genes showed a higher expression in follicles developed in the presence of FSH. However, lower doses of FSH demonstrated a stimulatory effect on AMH and BMP2 expression, while expression started to decline at the maximum dose. In this study, we have provided a better understanding of the mechanisms regulating AMH, AMHR II, and BMP2 signaling in GCs during folliculogenesis, which would improve the outcomes of conventional assisted reproductive technologies (ARTs), such as superovulation and oestrus synchronization in bovines.

Study on the expression patterns and function of JUNO and CD9 in bovine oocytes during in vitro maturation

Summary: A study investigated the expression patterns and functions of fertilization proteins JUNO and CD9 in bovine oocytes during in vitro maturation (IVM) and in vitro fertilization (IVF). The results showed that the mRNA and protein expression levels of JUNO and CD9 genes increased from bovine GV oocytes to MII oocytes, and similar expression patterns were observed in cumulus cells. Treatment with antibodies against JUNO or CD9 significantly reduced the fertilization rates of oocytes, with JUNO antibody showing a major impact on the fertilization ability of bovine oocytes.

Genetic Polymorphism and mRNA Expression Studies Reveal IL6R and LEPR Gene Associations with Reproductive Traits in Chinese Holsteins

Summary: In this study, we investigated the polymorphism effects of two metabolic-syndrome pathway genes, IL6R and LEPR, on reproductive performance in dairy cows. Through population-level association analyses and cell-level gene expression analyses, we found that several single nucleotide polymorphisms in IL6R and LEPR were significantly associated with reproductive traits in Chinese Holstein cows. The functional validation of these genes improved our understanding of cattle reproduction and provided molecular markers for genetic selection of reproductive traits in high-yielding dairy cattle.

Analysis of CircRNA Expression in Peripheral Blood of Holstein Cows in Response to Heat Stress

Summary: The study aimed to identify key circRNAs and pathways associated with heat stress in Holstein cows. By comparing milk yield, rectal temperature, and respiratory rate between heat stress and non-heat stress conditions, differences were observed indicating heat stress in cows. RNA-seq analysis revealed differentially expressed circRNAs enriched in multiple signaling pathways. Key circRNAs were identified as potential regulators of heat stress response in cows.

Dynamic changes of the gut microbial colonization in preterm infants with different time points after birth

Summary: Risks associated with preterm birth are unevenly distributed across all gestations, with complications such as NEC and LOS conditions more common at earlier gestational ages. The colonization process of fecal microbiota in preterm infants is highly dynamic at different time points after birth. Certain bacterial groups like Exiguobacterium, Acinetobacter, and Citrobacter decline in abundance with age, while Enterococcus (Klebsiella and Escherichia coli) gradually become the dominant microbiota at 42 days. Bifidobacteria colonization in preterm infants is relatively late, and the presence of the Chryseobacterium bacterial group varies at different time points.

Extraction and Elevation of Cell-Free DNA under Mastitis and Heat Stress in Dairy Cattle

Combination of CNP, MT and FLI during IVM Significantly Improved the Quality and Development Abilities of Bovine Oocytes and IVF-Derived Embryos

Summary: Oocyte maturation is crucial for female gametogenesis and subsequent fertilization and embryogenesis. This study investigated the effect of Pre-IVM with CNP, MT, and FLI on the developmental potential of bovine oocytes during in vitro maturation (IVM). The combination of CNP, MT, and FLI significantly improved the development of oocytes to blastocyst stage, as well as ATP content, GSH levels, TZP intensity, Delta psi m, cacline-AM fluorescence intensity, and reduced ROS levels. The survival and hatched rates after vitrification were also significantly higher in the CNP+MT+FLI group. Overall, targeting the combination of CNP, MT, and FLI can enhance the quality and developmental potential of bovine oocytes.

Summary: Genetic selection for immune response features is an effective strategy for improving animal health. This study identified differentially expressed genes (DEGs) associated with immune responses in dairy bulls, and the results revealed a relationship between DEGs and locomotion, tissue development, immune response, and detoxification. The KEGG pathway analysis showed that most DEGs were enriched in pathways related to disease, inflammation, and immune response, providing insights for genomic selection.

Phenotypic and Genetic Analyses of In Vitro Embryo Production Traits in Chinese Holstein Cattle

Summary: This study analyzed the phenotypic and genetic characteristics of in vitro embryo production traits in Chinese Holstein cattle, and investigated the factors influencing the production efficiency of in vitro embryo production. The results showed that the traits were significantly influenced by season and donor age, and exhibited heritability with low to high estimates. The count traits and proportion traits reflected different aspects of embryo production and should be considered in genetic selection to improve efficiency.
