Social and epidemiological aspects of dengue were evaluated in an important metropolitan area in southern Brazil, from August 2012 to September 2014. Demographic, clinical, serological data were collected from patients with acute dengue symptoms treated at public health system units (HSUs). A systematic approach to analyze the spatial and temporal distribution of cases was developed, considering the temporal cross-correlation between dengue and weather, and the spatial correlation between dengue and income over the city's census tracts. From the 878 patients with suggestive symptoms, 249 were diagnosed as positive dengue infection (28%). Considering the most statistically significant census tracts, a negative correlation was found between mean income and dengue (r = -0.65; p = 0.02; 95% CI: -0.03 to -0.91). The occurrence of dengue followed a seasonal distribution, and it was found to be three and four months delayed in relation to precipitation and temperature, respectively. Unexpectedly, the occurrence of symptomatic patients without dengue infection followed the same seasonal distribution, however its spatial distribution did not correlate with income. Through this methodology, we have found evidence that suggests a relation between dengue and poverty, which enriches the debate in the literature and sheds light on an extremely relevant socioeconomic and public health issue.

Lyophilization of Molecular Biology Reactions: A Review

Nayra Oliveira Prado, Larissa Araujo Lalli, Lucas Blanes, Dalila Luciola Zanette, Mateus Nobrega Aoki

Summary: Molecular biology is a widely used technique for research and diagnostics, involving the analysis of genetic material. Lyophilization, a dehydration process, can be applied to preserve enzymes, buffers, and reagents with specific storage conditions. This review presents a synthesis of the lyophilization process, describing each step and providing general information. Lyophilization protocols from the literature are also presented in tables for easy access by researchers seeking material for new protocols.

Ana Paula Muraro, Roseany Rocha, Alexandra Crispim Boing, Ligia Regina de Oliveira, Francine Nesello Melanda, Amanda Cristina de Souza Andrade

Summary: This study aims to examine the scale and characteristics of deaths caused by post-COVID conditions in Brazil. It is a descriptive study based on preliminary data from the 2021 Mortality Information System. The analysis considered records with ICD code B94.8 as the Basic Cause and code U09 in some lines of part I or II of the declaration. The study evaluated the distribution of deaths by geographic region, semester of occurrence, sex, age group, ethnicity/skin color, schooling, and place of occurrence. The findings revealed that there were 2,948 recorded deaths from post-COVID conditions, with varying rates across different regions in Brazil. More than half of the deaths occurred among males, individuals aged 60 years or older, and white individuals. The study concludes that deaths from post-COVID conditions have distinct sociodemographic characteristics across different regions.

Summary: Pancreatic cancer is a deadly disease with few known risk factors and susceptibility loci. The rarity of the disease and lack of adequate research power have hindered progress in understanding its genetic mechanisms. However, the PANDoRA consortium, the largest in Europe, has made significant contributions in discovering susceptibility loci, which will be crucial in developing preventive strategies.

Summary: Long noncoding RNAs (lncRNAs) are a type of RNA molecule that do not code for proteins and have a regulatory role. This study identified lncRNA-SNPs with a potential biological role in the development of breast cancer (BC) in the Brazilian population. Certain SNPs were found to be associated with BC development and patient characteristics.

Summary: Hepatitis B virus (HBV) is a chronic infection affecting approximately 300 million people, with current treatments being ineffective. The virus increases the risk of liver cirrhosis and hepatocellular carcinoma, causing nearly 1 million deaths annually. A new method using synthetic RNA to initiate HBV replication is described, eliminating background signals from input virus or plasmid DNA. This approach can be used to study multiple stages of HBV replication and identify sequence variants that confer resistance to antiviral drugs.

Summary: The lack of reliable early diagnostic tools is a major challenge in managing pancreatic cancer, which is often diagnosed at an advanced stage. The emergence of liquid biopsy as a less invasive approach focusing on plasmatic biomarkers such as DNA and RNA has addressed this issue. Circulating tumor cells (CTCs) and cell-free nucleic acids (cfNAs) like DNA, mRNA, and non-coding RNA (miRNA and lncRNA) in the blood of cancer patients have been investigated as potential biomarkers. This article focuses on circulating cfNAs as plasmatic biomarkers for pancreatic cancer and analyzes their advantages compared to traditional biopsy methods.

Summary: A study examined the composition of plasma virome in patients with prostate cancer, finding that Anelloviruses and Human Pegivirus-1 (HPgV-1) were the most abundant components. Clinically important viruses like hepatitis C virus (HCV), cytomegalovirus, and human adenovirus type C were also identified. In comparison, the virome of blood donors was mainly composed of torque teno virus types (TTV). This study sheds light on the viral abundance and diversity in patients with prostate cancer, suggesting that malignant processes may influence virome composition.
