Summary: Metabolic dysregulation in breast cancer was characterized and divided into two major metabolic groups. Genes strongly correlated with metabolic dysregulation and predicted patient prognosis were identified. Targeting metabolic dysregulation led to effective therapeutic outcomes. This study is of great significance in guiding therapeutic strategies for breast cancer subsets.

Summary: This study reveals the metabolic dependency induced by mutant KRAS and identifies nucleolar protein 5A (NOP56) and mTOR as potential targets for the treatment of KRAS-mutant cancers. The depletion of NOP56 leads to synthetic lethal susceptibility to mTOR inhibition due to the regulation of ROS homeostasis. Co-targeting NOP56 and mTOR significantly enhances cell death in KRAS-mutant tumor cells.

Summary: Cutaneous Melanoma (CM) is an aggressive cancer originating from pigment-producing melanocytes in the skin, with high metastatic potential. Although it represents a small fraction of skin cancers, it accounts for most skin-cancer-related deaths globally. Immune checkpoint inhibition has been a significant therapeutic approach, but challenges such as the immunosuppressive tumor microenvironment and treatment resistance persist. Combining local and systemic treatments can enhance therapeutic response and overcome resistance, yet complex drug combinations may increase the risk of immune-related adverse events.

Summary: Uveal melanoma (UM) is a common and dangerous ocular malignancy in adults, and aberrant histone methylation plays a role in its development. This study identified that DOT1L, a methyltransferase of histone H3 lysine 79 (H3K79), was activated in UM, especially in high-risk cases. DOT1L inhibitors showed tumor-selective inhibitory effects on UM cells. The study also revealed that DOT1L facilitated H3K79 methylation of NAPRT, which enhanced NAD+ synthesis and contributed to oncogenesis. DOT1L inhibition silenced NAPRT expression and inhibited the malignant behaviors of UM. These findings provide insights into the histone methylation landscape in UM and uncover a novel oncogenic mechanism involving DOT1L and NAPRT.

Summary: Satellite imagery from 2000 to 2018 indicates that the proportion of global population exposed to floods is increasing, highlighting the importance of investing in flood adaptation strategies to reduce loss of life and livelihood. Changes in where and how floods occur, as well as the population exposed, are being influenced by urbanization, flood mitigation infrastructure, and settlements in floodplains. The high spatial and temporal resolution of satellite observations will enhance understanding of flood changes and adaptation methods.
