Therefore, SLC25A32 can serve as an independent prognostic factor in GBM patients, providing a new target for the comprehensive treatment of GBM.

Summary: Obesity is a growing disorder that damages patients' quality of life and increases healthcare costs. Developing safe and innovative prevention strategies and treatments for obesity is crucial. The pathophysiology of obesity is complex and involves multiple signaling pathways, including the PI3K/AKT pathway, which plays a critical role in metabolic homeostasis. Natural compounds have been shown to target the PI3K/AKT signaling pathway and have potential for managing obesity. This paper evaluates the druggability of PI3K as a target and suggests perspectives on using natural compounds to modulate the PI3K/AKT pathway for obesity management.

Summary: Mulberroside A (MA), a natural bioactive withanolide, exhibits anti-inflammatory properties and protects articular cartilage in osteoarthritis (OA). In vitro, MA improves the inflammatory response, anabolism, and catabolism in OA chondrocytes. This chondroprotective effect is attributed to the suppression of MAPK, NF-kB, and PI3K-AKT-mTOR signaling pathways, as well as the promotion of autophagy. In vivo, intra-articular injection of MA reduces cartilage destruction and reverses the changes in anabolic and catabolic-related proteins in OA models. Therefore, MA may be a promising agent for OA treatment.

Summary: This study explored the pharmacological actions and mechanisms of Raddeanin A (RA) in non-small-cell lung cancer (NSCLC) using network pharmacology. The potential targets of RA for NSCLC therapy, such as SRC, MAPK1, and STAT3, were identified. Enrichment analyses indicated the involvement of these targets in regulating cell death, MAPK cascade, Ras signaling pathway, and PI3K/AKT signaling pathway. Experimental results showed that RA effectively inhibited proliferation, induced apoptosis, and induced autophagy in A549 lung cancer cells and downregulated the activity of the PI3K/AKT/mTOR pathway. Overall, this study revealed the antitumor effects and underlying mechanisms of RA on apoptosis and autophagy in A549 cells, suggesting its potential as an effective antineoplastic agent.

Summary: Euphorbia fischeriana is a traditional Chinese herb used for cancer treatment, insect repellent, anti-inflammatory, and anti-edema applications. 12-Deoxyphorbol13-palmitate (DP) extracted from this herb inhibited leukemia cell proliferation and induced apoptosis in vitro by suppressing the PI3k/Akt signaling pathways. In vivo experiments on nude mice showed significant tumor volume reduction in DP-treated groups, confirming its potential as an anti-leukemia agent.
