Imaging the spatial distributions and dynamics of flavones in heterogeneous plant tissues is significant for our understanding of plant metabolism. Here, we proposed a spatially-resolved approach to map the locations and biosynthesis of flavones in S. baicalensis. A total of 11 flavones, 5 flavone glycosides, 6 carbohydrates, and a variety of flavone synthesis-related metabolites were imaged. Most of these flavone-related metabolites presented stronger ion intensities in root phloem. The biosynthetic network of flavones and their glycosides in S. baicalensis were visualized for the first time. Moreover, we characterized the region-specific activities of four crucial enzymes in flavone synthesis pathway, including L-phenylalanine ammonia-lyase, cinnamate 4-hydroxylase, 4-coumarate coenzyme A ligase, and flavone synthase. In line with the spatial characteristic of flavones, all these four enzymes exhibit higher activity in the root phloem of S. baicalensis. The combination of spatially-resolved metabolites and enzymes information greatly broadens our understanding of flavone biosynthetic network. (C) 2019 Elsevier B.V. All rights reserved.

Summary: This study aimed to develop an efficient method for the extraction and separation of flavonoids from Gleditsia sinensis thorns using choline chloride deep eutectic solvents (DES) and high-speed counter-current chromatography (HSCCC). Five flavonoids were successfully obtained using this method, demonstrating that the combination of DES and HSCCC is a powerful technique for extracting and isolating flavonoids compared to traditional methods.

A novel on-tissue cycloaddition reagent for mass spectrometry imaging of lipid C=C position isomers in biological tissues

Summary: The application of matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) to investigate the changes of lipids in biological tissues has yielded significant results. However, the presence of structural isomers with varying C=C double bond (DB) locations calls for the development of isomer-resolved MSI. In this study, a new light-driven on-tissue [2 + 2] cycloaddition reaction coupled with MALDI-MS/MS imaging was introduced to identify lipid DB position isomers and their spatial signatures in biological tissues.

Comprehensive Lipidome and Metabolome Profiling Investigations of Panax quinquefolius and Application in Different Growing Regions Using Liquid Chromatography Coupled with Mass Spectrometry

Visualizing the spatial distribution and alteration of metabolites in continuously cropped Salvia miltiorrhiza Bge using MALDI-MSI

Summary: This study investigated the spatial location and variation of endogenous metabolites in continuously cropped Salvia miltiorrhiza Bge (SMB) roots using matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). The results showed that continuous cropping led to lower expression levels of tanshinones, salvianolic acids, polyamines, and oligosaccharides in SMB roots. However, there was little difference in the expressions of caffeic acid and salvianolic acid A. Ferulic acid was more widely distributed in continuously cropped SMB.

Multi-wavelength visible-light induced [2+2] cycloaddition for identification of lipid isomers in biological samples

Summary: A novel visible-light induced [2+2] cycloaddition reaction combined with mass spectrometry techniques was proposed to identify lipid C=C position isomers. The new cycloaddition reagents achieved high conversion efficiency under compact fluorescent lamp light, offering new insights into photocatalytic reagent development for visible-light induced cycloaddition and structural lipidomic studies.

Summary: The hybrid nanocomposite of gold nanoparticles and nickel/iron-layered double hydroxides can improve the performance of laser desorption/ionization mass spectrometry for low-molecular-weight samples, demonstrating high peak intensity, low background noise, salt and protein tolerance, good repeatability, and quantification. This nanocomposite also shows potential for detecting a variety of small molecules, providing rapid and accurate analysis in water samples.

Summary: In this study, a method based on MALDI-MS was developed for on-tissue derivatization, imaging, and quantification of aldose and ketose isomers of monosaccharides in biological tissues. The use of the newly synthesized derivatization reagent NAH significantly enhanced the imaging sensitivity of monosaccharides and allowed for isomer-specific MS imaging through the generation of isomer-specific diagnostic ions.

Summary: Matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) was used to characterize the expressions of fatty acids in breast cancer tissues, revealing that most fatty acids are up-regulated compared to adjacent normal tissues. Up-regulation of fatty acid synthase (FASN) and acetyl CoA carboxylase (ACC) was found in breast cancer, which can be targeted to limit cancer cell growth, proliferation, and metastasis.

Summary: In this study, a 3D tumor-immune cell coculture spheroid model combined with mass spectrometry imaging was developed to visualize the metabolic interactions between tumor and immune cells during immune response. The transport and catabolism of glutamine were found to be significantly reprogrammed during the antitumor immune response, indicating their important roles in energy metabolism and new biomass building. This integrated approach provides insights into the immunometabolic alterations in tumor immunotherapy.

A strategy takes Yiqing tablets as an example to carry out simpler multi-component quantification and use fingerprint technology for comprehensive quality consistency evaluation

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

Green sample preparation of medicinal herbs in closed digester block for elemental determination by ICP OES

Summary: This work presents a simple, cost-effective, and environmentally friendly method for determining the content of multiple elements in medicinal herbs using ICP OES. The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

Separation and characterization of related substances of Lurasidone hydrochloride by LC-QTOF-MS techniques

Summary: In this study, a reliable LC-QTOF-MS method was developed to separate and characterize process-related substances and degradation products of Lurasidone hydrochloride. The results showed that Lurasidone hydrochloride exhibited instability under certain stress conditions, and several previously unidentified compounds were discovered. The study emphasizes the importance of monitoring and controlling related substances in the manufacturing processes to optimize the process and ensure quality control.

The results showed that PEG600, PEG2K, PEG4K and PEG10K exhibited low toxicity and had limited cellular entry at concentrations of 5-1200 μg/mL. Additionally, the environmental friendliness of the developed analytical methods was evaluated.

Summary: This study employed network pharmacology-based analysis and reverse molecular docking to investigate the molecular targets and pathways associated with gynecological disorders and the potential therapeutic effects of fenugreek constituents. The results highlighted the potential of steroidal saponins, isoflavones, and flavones as active constituents of fenugreek with implications for lactation, reproductive processes, and estrogenic activity. Animal model study further supported the presence of these bioactive compounds and their effects on body weight, serum prolactin levels, serum estradiol levels, and ovarian histological structure.
