Hydroxytyrosol: A natural compound with promising pharmacological activities

Hydroxytyrosol is a phenolic phytochemical with antioxidant properties in vitro. It is a natural compound that can be found in olive leaves and oil. The main dietary source of hydroxytyrosol is extra virgin olive oil. Due to its bioavailability, chemical properties and easy formulation along with its lack of toxicity, hydroxytyrosol is considered an excellent food supplement by the nutraceutical and food industries. The purpose of this review is to discuss the potential therapeutic effects of hydroxytyrosol in vivo. To do so, we conducted an electronic search in PubMed and other literature databases using hydroxytyrosol, beneficial effect/s, pharmacology as keywords. From this search, we found that hydroxytyrosol has anti-inflammatory, anti-tumor, antiviral, antibacterial and antifungal properties. Hydroxytyrosol also improves endothelial dysfunction, decreases oxidative stress, and is neuro- and cardio-protective. Due to all these biological properties, hydroxytyrosol is currently the most actively investigated natural phenol. The evidence presented in this review suggests that hydroxytyrosol has great pharmacological potential.

A unique case associated with a de novo translocation affecting the KCNJ2 locus was reported, suggesting a complex rearrangement in this region may mimic the developmental consequences of in tandem duplications affecting the SOX9-KCNJ2 locus.

Useful molecular tools for facing next pandemic events: Effective sample preparation and improved RT-PCR for highly sensitive detection of SARS-CoV-2 in wastewater environment

Summary: This study selected an effective sample preparation procedure and combined it with a newly developed improved RT-PCR to detect viruses in wastewater samples. A model viral system was constructed using gene editing technology to evaluate the effectiveness of four viral RNA concentration/extraction methods. The research found that for wastewater samples with low viral loads, the OSN-qRT-PCR detection method performed the best and had the highest sensitivity.

Summary: The use of third generation sequencing technologies allows for longer sequence reads, making it possible to use the longest amplicons as specific bacterial markers for metabarcoding studies. Nanopore sequencing technologies are increasingly used worldwide to analyze microbiomes in environmental and food samples. This study optimized a PCR-based method for sequencing almost the entire ribosomal operon sequences and applied it to the analysis of bacterial consortia in dairy fermented products.

Unraveling disparate roles of organisms, from plants to bacteria, and viruses on built cultural heritage

Summary: The organisms inhabiting built cultural heritage can passively or actively participate in conservation processes. Active participants are focused on for their positive roles in various processes, as well as providing ecosystem services. These organisms can also evolve or mutate in response to changes and acquire new capacities. Recent studies have opened up new avenues of research involving a wide variety of organisms and viruses, which are expected to gain more attention in the future.

Summary: Manganese oxides are crucial in natural geochemical barriers, as they have reactive properties towards essential and potentially toxic trace elements. Microorganisms can actively alter conditions in their microenvironment and dissolve minerals, as well as precipitate bioavailable manganese ions into biogenic minerals. These microbial transformations significantly impact the geochemistry of manganese and elements associated with its oxides, potentially leading to environmental consequences.

Biodegradable Active Packaging Enriched with Essential Oils for Enhancing the Shelf Life of Strawberries

Summary: This study focuses on encapsulating essential oils (lemongrass and oregano) into PLA and PHB packaging films and evaluating their antimicrobial and antioxidant properties for strawberry preservation and shelf-life extension. The results show that the tested active packaging film with essential oils can effectively maintain the quality and extend the shelf life of strawberries, with PLA/PHB/ATBC + 5% lemongrass essential oil performing slightly better than PLA/PHB/ATBC + 5% oregano essential oil.

Summary: This study investigated the age-related changes and central retinal impairment in patients with EYS gene mutations. It found a positive correlation between RP stage scoring system (RP-SSS) and age, as well as a correlation between RP-SSS and central retina atrophy (CRA) area. Additionally, visual acuity and ellipsoid zone width were correlated with CRA, but not electroretinogram (ERG).

Development of Salmon Sperm DNA/Regenerated Silk Bio-Based Films for Biomedical Studies on Human Keratinocyte HaCaT Cells under Solar Spectrum

Summary: In this study, adhesive patches made of silkworm-regenerated silk and DNA were developed to protect human skin from the sun's rays. The addition of DNA increased the crystallinity of silk fibroin (SF), as observed through infrared spectroscopy. UV-Visible absorption and circular dichroism spectroscopy confirmed the strong UV absorption and presence of B-form DNA in the SF matrix. Biological results showed that the SF/DNA patches increased the viability of keratinocytes after UV exposure. These patches have promising applications in wound dressing and biomedical purposes.

MAGI-ACMG: Algorithm for the Classification of Variants According to ACMG and ACGS Recommendations

Summary: We have developed MAGI-ACMG, a classification algorithm that allows the classification of sequencing variants according to the recommendations of ACMG and ACGS. This algorithm utilizes the VarSome API, integrates the AutoPVS1 tool for precise evaluation, and performs customized assignment of criteria. Additionally, we propose a sub-classification scheme for variants of uncertain significance and introduce a pathogenicity potential criterion for upgrading clinical significance.

Summary: Wastewater monitoring is an important approach for detecting and controlling the SARS-CoV-2 pandemic. In this study, a one-step single-tube nested qRT-PCR assay was developed to increase the sensitivity of qRT-PCR. The assay demonstrated higher detection rates and robustness in samples with low viral loads.

Summary: This study aimed to increase knowledge of genes associated with anorexia nervosa (AN) and their diagnostic offer using next generation sequencing (NGS). They found potentially deleterious variants in genes associated with syndromic forms of anorexia and identified candidate genes playing a possible causative role in AN etiology through family segregation studies. The study confirmed the involvement of the NNAT gene in the pathogenesis of AN and highlighted the importance of NGS-based testing in the diagnostic evaluation of AN.
