Summary: Neutrophil migration into the lung is mediated by cell adhesion molecules such as ICAM-1 and ICAM-2, which are upregulated during inflammatory stimulation. While beta(2) integrins interacting with ICAMs may regulate neutrophil traffic in healthy lung, these interactions may become redundant later in inflammation when epithelial junctions are disrupted.

Summary: We have reported a series of compounds that have the potential to manage oxaliplatin-induced neuropathy (OINP) by modulating human Carbonic Anhydrases (hCAs) and Transient Receptor Potential Vanilloid 1 (TRPV1). These compounds showed effective inhibition activity against the main hCAs involved in OINP in vitro, and some of them exhibited moderate agonism of TRPV1. In vivo evaluation of promising derivatives demonstrated potent and persistent antihypersensitivity effects in a mouse model of OINP.

Summary: The study demonstrates that RUT promotes NO synthesis and eNOS phosphorylation by increasing intracellular Ca2+ concentration and kinase phosphorylation. Inhibition of TRPV1 attenuates RUT-induced effects, and blocking downstream signaling pathways suppresses eNOS phosphorylation and NO generation. These findings indicate that RUT can prevent endothelial dysfunction and benefit cardiovascular health through specific signaling pathways.

Summary: Using a photoconvertible probe, researchers traced and identified the original endothelial transmigration hotspots, and found that these hotspots function to limit vascular leakage during transmigration events. The distribution of ICAM-1 determines the location of the transmigration hotspot.

Summary: Colorectal cancer is a major global health problem, and understanding the role of intercellular adhesion molecule-1 (ICAM-1) in its pathogenesis is crucial for early diagnosis, prevention strategies, and identification of therapeutic targets. ICAM-1 plays a significant role in the interaction between cells, cell signaling, and immune processes. Its expression in tumor cells modulates their functions and may contribute to the adhesion and metastasis of cancer cells. This review discusses the expression, function, prognosis, tumorigenesis, polymorphisms, and therapeutic implications of ICAM-1 in colorectal cancer.

Summary: This study demonstrates that astrocytes produce endogenous CNTF through TRPV1-p70S6K signaling, which plays a neuroprotective role in Parkinson's disease. The findings suggest that targeting astrocytic TRPV1-p70S6K signaling may be a novel therapeutic strategy for PD.

Summary: The study reveals that endothelial ADAM10 plays a role in facilitating neutrophil transendothelial migration by cleaving ICAM-1, supporting the efficient release of neutrophils from the endothelium. Silencing or inhibiting endothelial ADAM10 leads to a significantly longer diapedesis step for neutrophils due to impaired dissociation from ICAM-1. Increased ICAM-1 levels on transmigrating neutrophils and potential membrane transfer from endothelium to neutrophils are also observed.

Summary: Leukocyte migration is vital for lymphocyte recirculation and immune response. ICAM-1 and VCAM-1 play a crucial role in leukocyte adherence to endothelial cells in various diseases. They contribute to inflammation, leukocyte migration, and have implications in multiple pathological states. Understanding their immunoregulatory role and pathophysiological effects helps in disease management.

Summary: TRPV1 is overexpressed in non-small-cell lung cancer, and its overexpression leads to increased resistance to DDP and 5-FU. TRPV1 mediates chemoresistance by increasing drug efflux, enhancing DNA repair pathways, and promoting cell survival.

Summary: This study reported the generation and characterization of ICAM-1-specific nanobodies, which showed high affinity binding to ICAM-1 and were not internalized by cells. The findings suggest that these nanobodies can be used in combination with allergen-specific nanobodies to develop topical treatments for pollen allergy.
