Aquaglyceroporins Are Differentially Expressed in Beige and White Adipocytes

Browning of white adipocytes has been proposed as a powerful strategy to overcome metabolic complications, since brown adipocytes are more catabolic, expending energy as a heat form. However, the biological pathways involved in the browning process are still unclear. Aquaglyceroporins are a sub-class of aquaporin water channels that also permeate glycerol and are involved in body energy homeostasis. In the adipose tissue, aquaporin-7 (AQP7) is the most representative isoform, being crucial for white adipocyte fully differentiation and glycerol metabolism. The altered expression of AQP7 is involved in the onset of obesity and metabolic disorders. Herein, we investigated if aquaglyceroporins are implicated in beige adipocyte differentiation, similar to white cells. Thus, we optimized a protocol of murine 3T3-L1 preadipocytes browning that displayed increased beige and decreased white adipose tissue features at both gene and protein levels and evaluated aquaporin expression patterns along the differentiation process together with cellular lipid content. Our results revealed that AQP7 and aquaporin-9 (AQP9) expression was downregulated throughout beige adipocyte differentiation compared to white differentiation, which may be related to the beige physiological role of heat production from oxidative metabolism, contrasting with the anabolic/catabolic lipid metabolism requiring glycerol gateways occurring in white adipose cells.

Mitochondrial dysfunction and alterations in mitophagy induce inflammatory responses, but whether accumulation of damaged mitochondria in muscle triggers inflammation during aging is still unknown.

Summary: This study investigated the association between circulating NRG4 and insulin sensitivity in humans. The results showed a negative correlation between serum NRG4 and insulin sensitivity, and a positive correlation with the inflammatory marker hsCRP. In vitro experiments also revealed that NRG4 had an impact on hepatocyte metabolism. However, further confirmation is needed in human primary hepatocytes.

Disruption of mitochondrial dynamics triggers muscle inflammation through interorganellar contacts and mitochondrial DNA mislocation

Summary: Some forms of mitochondrial dysfunction induce sterile inflammation through mitochondrial DNA recognition by intracellular DNA sensors. Opposite mitochondrial morphologies induce distinct inflammatory signatures, caused by differential activation of DNA sensors TLR9 or cGAS. Skeletal muscle mitochondrial fragmentation promotes TLR9-dependent inflammation, muscle atrophy, reduced physical performance and enhanced IL6 response to exercise, which improved upon chronic anti-inflammatory treatment.

Unraveling the Aquaporin-3 Inhibitory Effect of Rottlerin by Experimental and Computational Approaches

Summary: The natural polyphenolic compound Rottlerin (RoT) has shown potential as an anticancer agent by inhibiting target molecules implicated in tumorigenesis. It has been found to inhibit human AQP3 activity with an IC50 in the micromolar range. Molecular docking and simulations have revealed the structural determinants of RoT that explain its ability to inhibit AQP3. This multidisciplinary approach provides new information for aquaporin research and future drug design.

AQP3-Dependent PI3K/Akt Modulation in Breast Cancer Cells

Summary: Aquaporin 3 (AQP3) is a membrane protein that transports hydrogen peroxide, water, and glycerol. It is implicated in tumor progression and malignancy in breast cancer and may be a target for therapy. This study examined the relationship between AQP3 expression, disruption of lipid raft harboring EGFR, and the EGFR/PI3K/Akt signaling pathway in breast cancer cell lines. The results indicate that AQP3 may not directly affect the PI3K/Akt pathway but rather its role is cell-line specific.

Summary: This study investigated the potential effect of a dietary amino acid mixture on LPS-challenged weaned piglets and found that the amino acid mixture may contribute to immune recovery. Further analysis revealed tissue-specific variation in the gene expression of AQPs and cytokines, distinguishing the small intestine and kidney from the liver and spleen.

Summary: We investigated the role of AQP7 in the process of BAT whitening and found that its expression is upregulated in diet-induced obesity (DIO). Interestingly, the expression of AQP7 is downregulated after cold exposure or bariatric surgery, which is associated with the improvement of BAT whitening. Furthermore, AQP7 expression is regulated by specific lipogenic signals and correlated with the expression of lipogenic genes.
