N-acetylglucosaminidase produced from Lecanicillium lecanii on submerged culture displayed hydrolytic and transglycosylation activities. The highest specific activity for the enzyme was 1.87 U/mg after 120 h of culture. The chromatographic purification for a single protein fraction showed a molecular weight of 50.4 kDa and hydrolytic N-acetylglucosaminidase activity of 17.59 U/mg at 37 degrees C and pH 6. This enzyme was able to transglycosylate and to synthesize oligosaccharides from 2 to 6 units with a degree of acetylation between 100 and 26% employing glucose, mannose, N-acetyl-D-glucosamine and N-acetyl-D-lactosamine as donor substrates. Optimal conditions of temperature and pH were determined for both types of enzymatic activities.

Summary: Proteomic techniques were used to identify the most abundant proteins in the venom of Chrysaora chesapeakei from the Gulf of Mexico estuaries, revealing main toxins including metalloproteinase-zinc disintegrin and astacin-type metalloprotease. These components could be responsible for the main effects of this jellyfish venom. Additionally, nonpoisonous metalloproteinases were found in the connective tissue of the tentacles.

Population dynamics of nitrifying sludge supplemented with 2-chlorophenol in SBR reactors

Miguel Martinez-Jardines, Rosa O. Gonzalez-Robles, H. Gonzalez-Marquez, Anne-Claire Texier, Flor de Maria Cuervo-Lopez

Summary: The metabolic improvement and reduction of the inhibitory effect of 2-chlorophenol (2-CP) on the nitrification process occurring in nitrifying SBR systems can be associated with changes in the microbial community. The stable, complete, and efficient nitrifying process achieved in both SBRs could be related to the higher bacterial community richness and to the regular maintenance of ammonium and nitrite oxidizing bacteria tolerant to 2-CP along experimentation. Total 2-CP consumption might be associated with ammonium-oxidizing and/or heterotrophic bacteria related to Nitrosospira, Luteimonas, and Cohnella.

Perfluorooctane sulfonate and perfluorooctanoic acid induce plasma membrane dysfunction in boar spermatozoa during in vitro capacitation

Paola Berenice Ortiz-Sanchez, Ana L. Roa-Espitia, Reyna Fierro, Aidee S. Lopez-Torres, Irma Jimenez-Morales, Ivan Oseguera-Lopez, Enrique O. Hernandez-Gonzalez, Humberto Gonzalez-Marquez

Summary: This study examines the effects of PFOS and PFOA on boar spermatozoa functionality and changes in the plasma membrane during capacitation. The results indicate that both substances reduce the capacitation ability of spermatozoa and impede the progesterone-induced acrosomal reaction. PFOS and PFOA also cause changes in the plasma membrane, including deregulation of membrane potential and alteration of membrane microdomain organization.

Summary: Obesity is associated with male infertility due to increased oxidative stress on sperm. This study found that overweight and obesity led to lower testosterone levels and poor sperm quality, although fertility was not greatly affected. However, the increased activities of CAT and GPX enzymes seemed to protect sperm from oxidative stress by counteracting the increase in ROS.

Effects of aging and diabetes on the deformation mechanisms and molecular structural characteristics of collagen fibrils under daily activity

Wei-Han Hui, Yen-Lin Chen, Shu-Wei Chang

Summary: Crosslinking affects the mechanical behavior and tissue metabolism of collagen-based tissues. Aging and diabetes alter the crosslinking type and density, thus changing tissue properties. High-connectivity enzymatic crosslinking and advanced-glycation end-products result in uniform deformation under daily activity, while low-connectivity enzymatic crosslinking does not. High-connectivity model displays more sliding, while AGEs induce instability in structures near the binding sites.
