Lidocaine tripotassium phosphate complex laden microemulsion for prolonged local anaesthesia: In vitro and in vivo studies

Lidocaine is widely used as a local anaesthetic in the clinical practice to manage pre- and post-operative pain, skin burns, etc. However, the short duration of action ( < 2 h) of marketed dosage forms limit their ability to meet clinical needs. Herein, we prepared a lidocaine-tPP(tri potassium phosphate)-complex loaded micro emulsion to achieve greater penetration, followed by destabilization of microemulsion in the skin layer to precipitate oil -complex to produce a depot effect in the skin for prolonging the effects of anaesthesia. The lidocaine-tPP-complex-microemulsion was compared with lidocaine base loaded microemulsion, marketed ointment USP and lidocaine HCl. The pseudo ternary phase diagrams at three S-mix ratios (1:2, 1:3 and 1:4; Pluronic F127: PEG 400) were constructed using Capmul MCM C8 EP as oil phase. The S-mix at 1:4 ratio showed large microemulsion area in comparison to 1:2 and 1:6 ratio. The lidocaine base (LD-1:4-ME10O45SM and LD1:4-ME20O45SM) and lidocaine-tPP-complex (LDC-1:4-ME10O45SM and LDC-1:4-ME20O45SM) loaded microemulsion batches (1:4 ratio) were thermodynamically stable. The ex vivo diffusion study showed sustained release up to 12 h with microemulsion batches, in comparison to lidocaine HCl (4 h) and ointment base (7 h). The selected LDC-1:4-ME20O45SM batch was non-irritating on the rabbit skin. In drug retention studies, LD-1:4-ME20O45SM and LDC-1:4-ME20O45SM batches showed 2.68- and 3.93-fold greater lidocaine retention in comparison to ointment USP. The radiant heat tail-flick test showed prolong local anaesthesia using LDC-1:4ME20O45SM in comparison to ointment USP. The findings suggest that lidocaine-tPP-complex loaded micro emulsion could be a potential strategy for providing prolong local anaesthesia.

The method parameters were assessed for risk and optimized using a full-factorial design, ensuring control of critical parameters throughout the method's lifecycle. The RP-HPLC method was successfully validated and used for simultaneous estimation of telmisartan FDC products, meeting the labeled claim. This method fulfills the requirements of various RP-HPLC and high-performance thin layer chromatography methods.

Monosaccharide-induced growth and higher order transitions in TPGS micelles

Summary: This study demonstrates that D-alpha-tocopheryl polyethylene glycol 1000 succinate (TPGS) spherical micelles can transform into ellipsoid and cylindrical structures upon the addition of monosaccharides such as glucose, mannose, and galactose. Structural transitions are driven by sugar-induced dehydration in the head group region, with different sugars showing varying abilities to trigger these transitions. NMR experiments reveal structural reorganizations in TPGS molecule in the presence of sugars.

Antiviral Activity of Contemporary Contact Lens Care Solutions against Two Human Seasonal Coronavirus Strains

Summary: According to the principles of WAC, it is important to avoid using teratogenic and carcinogenic organic solvents in order to protect the environment and analysts. Therefore, a green RP-HPLC method based on WAC was developed and validated for the simultaneous analysis of anti-hypertensive drugs in human plasma. The development of the method was guided by the analytical quality by design approach and involved the optimization of critical method variables using design of experiments and chemometrics concepts.

Small molecule antiviral agents targeting HBV and HCV have advanced into different phases of clinical investigation, mainly targeting complex molecular interactions between virus and host cells involved in viral replication.

Graphene oxide-doped chiral dextro-hydrogel promotes peripheral nerve repair through M2 polarization of macrophages

Analysis of surfactant production by Bacillus cereus GX7 and optimization of fermentation conditions

Summary: In this study, a highly effective surfactant producer strain, Bacillus Cereus GX7, was isolated from the oil tank bottom sludge of Shengli Oil Field in China. The biosurfactant produced by GX7 was identified as surfactin, a lipopeptide surfactant, through TLC, FT-IR, and LC-MS/MS analysis. The fermentation process of GX7 was optimized using single-factor experiments, focusing on the composition of fermentation medium and fermentation conditions. Glucose and peptone were found to be the best carbon and nitrogen sources, and the optimum temperature, inoculum amount, pH, rotation speed, and fermentation time for the strain were determined to be 30°C, 1%, 7.5, 150 rpm, and 48 h, respectively. After optimization, the surface tension and emulsification index of the fermentation broth were 26.84 mN/m and 57.84%, respectively. Furthermore, the biosurfactant produced by GX7 demonstrated good stability over a wide range of temperature, pH, and salt concentration.

In this study, researchers developed an interpenetrating Janus wettability stent coating that enables unidirectional draining of excessive biofluid. They also demonstrated directional biofluid movement using a self-pumping dressing with potential applications in biofluid collection and disease diagnosis through metal ion detection. This integrated system presents an opportunity for designing wound dressings with effective biofluid management and metal ion detection capabilities.
