PDGFB-expressing mesenchymal stem cells improve human hematopoietic stem cell engraftment in immunodeficient mice

The bone marrow (BM) niche regulates multiple hematopoietic stem cell (HSC) processes. Clinical treatment for hematological malignancies by HSC transplantation often requires preconditioning via total body irradiation, which severely and irreversibly impairs the BM niche and HSC regeneration. Novel strategies are needed to enhance HSC regeneration in irradiated BM. We compared the effects of EGF, FGF2, and PDGFB on HSC regeneration using human mesenchymal stem cells (MSCs) that were transduced with these factors via lentiviral vectors. Among the above niche factors tested, MSCs transduced with PDGFB (PDGFB-MSCs) most significantly improved human HSC engraftment in immunodeficient mice. PDGFB-MSC-treated BM enhanced transplanted human HSC self-renewal in secondary transplantations more efficiently than GFP-transduced MSCs (GFP-MSCs). Gene set enrichment analysis showed increased antiapoptotic signaling in PDGFB-MSCs compared with GFP-MSCs. PDGFB-MSCs exhibited enhanced survival and expansion after transplantation, resulting in an enlarged humanized niche cell pool that provide a better humanized microenvironment to facilitate superior engraftment and proliferation of human hematopoietic cells. Our studies demonstrate the efficacy of PDGFB-MSCs in supporting human HSC engraftment.

By analyzing the dynamic changes of CRISPR-Cas9 system in iPSCs, new genes can be explored to improve knockin efficiency; reducing cell death and interfering with DNA damage repair signal may be important ways to improve efficiency when treating iPSCs.

Co-Mutation of FAT3 and LRP1B in Lung Adenocarcinoma Defines a Unique Subset Correlated With the Efficacy of Immunotherapy

Summary: Through comprehensive analysis of co-mutated genes in lung adenocarcinoma samples, the co-mutation of FAT3 and LRP1B is found to be associated with age and ethnicity in lung adenocarcinoma patients, and it can impact tumor burden, cytotoxicity, tumor immunogenicity, and lymphocyte infiltration. Additionally, the subset of LUAD with co-mutation of FAT3 and LRP1B exhibits significantly prolonged immunotherapy progression free survival.

Factor VII deficiency in China: Phenotype, genotype and current status of management

Summary: This study investigated the clinical phenotype and genotype of patients with congenital factor VII (FVII) deficiency in China and showed their current management status. The majority of patients had FVII activity <= 10%, which was associated with various bleeding symptoms. Prothrombin complex concentrates were the most frequently used on-demand replacement therapy. Prophylaxis before delivery decreased the risk of postpartum bleeding in women of reproductive age.

Clinical and molecular characteristics of forty Chinese children with essential thrombocythemia: A single-center, retrospective analysis

Summary: Little is known about the pathophysiological mechanism of essential thrombocythemia (ET) in children due to its rarity. A retrospective analysis of 40 Chinese children with ET from 2015-2021 revealed that more than half of them were asymptomatic at diagnosis. Microvascular symptoms were observed in nearly half of the children, while only two cases experienced vascular events. Driver gene mutations were found in a low proportion of children, and the most prevalent non-driver gene mutations were less common compared to adults with ET.

Molecular analysis of phenotypic heterogeneity in JAK2V617F-positive myeloproliferative neoplasms reveals a potential target for therapy

Summary: JAK2V617F is the most frequent mutation in BCR-ABL-negative myeloproliferative neoplasms (MPNs). Two concurrent mutations in SH2B3 and SF3B1 were identified, which may affect the phenotype of MPNs. Transcriptomic analysis revealed elevated inflammatory signaling pathways in ET HSCs and MEPs, and upregulation of Wnt/beta-catenin signaling during ET hematopoietic differentiation. Inhibition of Wnt/beta-catenin signaling decreased platelet counts in JAK2V617F(+) MPN mice. Overall, these findings provide insights into phenotypic differentiation of JAK2V617F(+) PV and ET and suggest Wnt/beta-catenin signaling as a potential therapeutic target for MPNs.

Women and girls with haemophilia: A retrospective cohort study in China

Summary: This study is the first and largest in China to describe women and girls with haemophilia. It found deficiencies in the identification, diagnosis, and management of these patients. Improving health insurance policies, establishing haemophilia centres, and creating multidisciplinary teams for bleeding and perinatal or perioperative management will help reduce morbidity and mortality.

Summary: This study compared the diagnostic utility of virtual bronchoscopic navigation (VBN) and radial endobronchial ultrasound with a guide sheath (rEBUS-GS) with or without fluoroscopy for transbronchial sampling of peripheral pulmonary lesions (PPLs). The results showed that the diagnostic yield was similar in both groups, and the group without fluoroscopy had shorter operation time and no severe complications.

Incidence and risk factors for second malignancies among patients with myeloproliferative neoplasms

Summary: This study retrospectively analyzed the clinical characteristics and survival of 1060 Chinese patients with myeloproliferative neoplasms (MPNs) to explore the risk factors for secondary cancer in MPN patients. The results showed that the survival of MPN patients with secondary cancer was significantly worse, and age >= 65 years and MF-1 were identified as risk factors for developing secondary cancer.

Summary: Researchers have developed a method to generate induced mesenchymal stromal cells (iMSCs) by directly reprogramming human peripheral blood mononuclear cells (PBMCs) using five factors. Omission of OCT4, one of the factors, significantly impairs the functionality of iMSCs and downregulates MSC-specific and mesoderm-regulating genes. The transient expression of OCT4 increases chromatin accessibility and promotes demethylation.
