Summary: This study investigated the clinical significance and molecular mechanisms of HMGA1 as a tumor-suppressing gene in hepatocellular carcinoma (HCC). The results showed that HMGA1 was significantly higher in HCC tissues and correlated with poor prognosis. Gene analysis revealed potential relationships between HMGA1 and other genes in HCC. Downregulation of HMGA1 suppressed HCC cell proliferation and migration, and blocked cell cycle progression. Further investigation identified miR-195-5p as a direct regulator of HMGA1, and its downregulation abolished the effects of HMGA1 on HCC. The study provides strong evidence for the role of HMGA1 in promoting HCC and its negative regulation by miR-195-5p.

Summary: This study reveals that TINCR is upregulated in HCC and associated with poor patient prognosis. Functional experiments show that silencing TINCR inhibits HCC proliferation, migration, invasion, and oxaliplatin resistance, while overexpressing TINCR has the opposite effects. Mechanistically, TINCR acts as a competing endogenous RNA to sponge miR-195-3p, relieving its repression on ST6GAL1 and activating nuclear factor kappa B signaling.

Summary: Long noncoding RNA MCF2L-AS1 plays a role in the development of various cancers, but its function in hepatocellular carcinoma (HCC) was unclear. This study investigated its role in HCC cell proliferation, migration, and invasion. MCF2L-AS1 was highly expressed in HCC tissues and cells, and its upregulation enhanced HCC cell behaviors while reducing apoptosis. miR-33a-5p was identified as a target of MCF2L-AS1 and inhibited HCC cell behaviors. The MCF2L-AS1-miR-33a-5p-FGF2 axis may be a potential therapeutic target for HCC treatment.

Summary: This study found that the expression of miR-6071 was decreased in liver cancer stem cells (LCSCs). Overexpression of miR-6071 inhibited self-renewal and tumorigenesis of LCSCs, as well as proliferation and migration of hepatocellular carcinoma (HCC) cells. The study also revealed that miR-6071 targeted the 3’UTR of PTPN11 mRNA and that upregulation of miR-6071 sensitized HCC cells to lenvatinib treatment. Clinical analysis showed that HCC patients with high miR-6071 expression had better survival outcomes with postoperative lenvatinib treatment.
