Development of a multiplex immunochromatographic strip test and ultrasensitive electrochemical immunosensor for hepatitis B virus screening

This research proposes two methods for hepatitis B diagnosis including rapid testing and electrochemical assay. For the first method, a multiplex hepatitis B test strip was fabricated to serve as a rapid test for hepatitis B screening. It was developed to simultaneously test three essential serological markers of hepatitis B virus infection including hepatitis B surface antigen (HBsAg), hepatitis B surface antibody (Anti-HBs) and hepatitis B core antibody (Anti-HBc). Gold nanoparticles (GNPs) were used as the signal generator on the test strip. Furthermore, a part of a paper network was incorporated on the strip for the gold-silver enhancement process. This paper network helped in decreasing the analysis time of enhancement and makes the enhancement process easier for rapid testing. The developed test strip was specific for each serological marker. The detection limits of HBsAg, Anti-HBs and Anti-HBc were obtained at 0.5, 0.3 and 0.1 mu g mL(-1), respectively. For the second method, electrochemical impedance spectroscopy (EIS) was applied for HBsAg detection. This method was proposed for quantitative hepatitis B detection. Anti-HBs antibodies were immobilized on a carbon screen printed electrode (SPCE) via the N-ethyl-N'-(3-(dimethylamino)propyl)carbo-diimide/N-hydroxy succinimide (EDC/NHS) couple reaction which reacted with the carboxyl group of the BSA cross-linked film on the electrode. The electrode modification process was characterized by EIS. A linear relationship between delta charge transfer resistance (Delta Rct) and HBsAg concentration was obtained in the range of 5-3000 ng mL(-1) with a detection limit of 2.1 ng mL(-1). This work is appropriate for quantitative analysis because it is a simple and low-cost method to implement as the SPCE is disposable. Therefore, we hope that this research will be useful to improve hepatitis B detection in the future. (C) 2019 Elsevier B.V. All rights reserved.

Disposable Electrochemical Sensor for Food Colorants Detection by Reduced Graphene Oxide and Methionine Film Modified Screen Printed Carbon Electrode

Summary: The study investigated the performance of Fe3O4 catalysts with Ni and Cr dopants in ethanol steam reforming reaction, with NiFe2O4 demonstrating the highest catalytic activity and stability for hydrogen production. The incorporation of Ni in the spinel ferrite structure enhanced reduction efficiency of iron compounds, improving catalytic activity and stability during the ESR process. Additionally, NiFe2O4 catalyst showed the ability to regenerate during the ESR process, as confirmed by X-ray absorption spectroscopy.

Encapsulation of α-tocopherol in large-ring cyclodextrin containing 26 α-D-glucopyranose units: A molecular dynamics study

Summary: By using molecular dynamics simulations, this study investigated the encapsulation of alpha-tocopherol with CD26, a supramolecular complex, and found that alpha-tocopherol spontaneously interacts with CD26 within 20 ns and forms a stable inclusion complex during 150-400 ns. The results showed that there were two groups of inclusion complexes, fully and partly encapsulated, which can enhance the solubility and stability of alpha-tocopherol.

Summary: The transparent nanocomposite sol of graphene quantum dot and amorphous TiO2 (GQD/a-TiO2) showed enhanced photocatalytic activity in degradation of RhB due to the interactions between surface functional groups on GQD sheets and a-TiO2, leading to band gap narrowing and synergistic effects. The smaller size of GQD facilitated greater extent of interactions at nanoscopic level, resulting in improved photocatalytic performance.

Synthesis and characterization of reduced graphene oxide/cobalt sulfide heterostructures as the visible-light-driven photocatalysts

A facile one-pot synthesis of magnetic iron oxide nanoparticles embed N-doped graphene modified magnetic screen printed electrode for electrochemical sensing of chloramphenicol and diethylstilbestrol

Summary: In this study, a simple one-pot solvothermal synthesis approach was used to fabricate a magnetic iron oxide embed nitrogen-doped graphene nanohybrid. This nanohybrid was then applied in an electrochemical sensor for the ultrasensitive detection of antibiotic residue contamination in milk samples. The sensor demonstrated excellent performance with high sensitivity, selectivity, and fast detection.

All-Atom Molecular Dynamics Simulations on a Single Chain of PET and PEV Polymers

Structural dynamics and kinase inhibitory activity of three generations of tyrosine kinase inhibitors against wild-type, L858R/T790M, and L858R/ T790M/C797S forms of EGFR

Summary: In this study, the effects of EGFR mutations on the susceptibility of different EGFR inhibitors were investigated using comprehensive molecular modeling and in vitro kinase inhibition assays. The results showed that three hot-spot residues were involved in the binding of osimertinib and afatinib to mutated EGFRs, enhancing their inhibitory activity. Furthermore, principal component analysis revealed that the molecular complexation of osimertinib against mutated EGFRs was in a closed conformation, leading to drug resistance.

Utilization of Receptor-Binding Domain of SARS-CoV-2 Spike Protein Expressed in Escherichia coli for the Development of Neutralizing Antibody Assay

Summary: This study explores the potential usefulness of E. coli as an alternative expression system for producing the receptor-binding domain (RBD) of SARS-CoV-2, which can be used for the development of a cost-effective and flexible neutralizing antibody detection assay. The results suggest that non-glycosylated RBD produced from E. coli may be a viable alternative to expensive glycosylated RBD produced from human cells in the development of such an assay.

The engineered Nbs show potential for the development of novel treatment, prevention, and diagnostic strategies against SARS-CoV-2.

Native top-down mass spectrometry for monitoring the rapid chymotrypsin catalyzed hydrolysis reaction

Summary: Enzymes play a crucial role as biological catalysts in accelerating biochemical reactions in living organisms. This study proposes a convenient method for monitoring enzymatic catalytic processes using native top-down mass spectrometry. By exploring the heterogeneity of the chymotrypsin sample and using tandem mass spectrometry, the researchers were able to monitor covalent and noncovalent enzymatic complexes, substrates, and products during catalysis. The results demonstrate that this method has the potential to be a promising tool for characterizing biocatalysts.

Carbon fiber-based multichannel solid-contact potentiometric ion sensors for real-time sweat electrolyte monitoring

Summary: This study presents a method for constructing wearable sweat electrolyte sensors using carbon fiber-based solid-contact ion-selective electrodes (SC-ISEs). By using carbon fibers extracted from commercial cloth as electrode material, the cost and reproducibility issues of flexible SC-ISEs were addressed. The results showed that the carbon fiber-based SC-ISEs exhibited reversible voltammetric and stable impedance performances, and had high reproducibility of standard potentials between normal and bending states.

Imaging peroxynitrite in endoplasmic reticulum stress and acute lung injury with a near-infrared fluorescent probe

Summary: A near-infrared fluorescent probe has been developed to accurately measure the levels of peroxynitrite (ONOO-) in the endoplasmic reticulum (ER) in acute lung injury (ALI). The probe demonstrated rapid response, high selectivity, good sensitivity, and enhanced fluorescence intensity in response to ONOO-. It was successfully used to detect changes in ONOO- levels and showed significant increases in an ALI cell model and an ALI mouse model.

Miniaturized ascorbic acid assay platform based on point discharge atomic emission spectrometry coupling with gold filament enrichment

Summary: In this work, a portable analytical system based on point discharge chemical vapor generation atomic emission spectrometry (PD-CVG-AES) coupling with gold filament enrichment was designed. The highly sensitive analysis of Hg2+ indirectly realized the detection of ascorbic acid (AA). The measurement is based on the fact that Ag+ can decrease the concentration of Hg2+ by forming Ag-Hg amalgam in the presence of the reductant SnCl2, while AA can pre-reduce Ag+ to Ag0, leading to the generation of silver nanoparticles (Ag NPs). The developed novel analytical strategy broadens the application of microplasma-based AES and offers a higher level of sensitivity compared to current AA detection techniques.

Time-gated luminescent probes for lysosomal singlet oxygen: Synthesis, characterizations and bioimaging applications

Summary: This study developed two lysosome-targetable background-free TGL probes for efficient and accurate detection of 1O2, which can be used for monitoring endogenous 1O2 concentrations in lysosomes and discriminating variability induced by different photosensitizers. Furthermore, a smart luminescent sensor film was successfully prepared for on-site 1O2 production detection during PDT processes, offering a promising clinical monitoring tool for skin diseases.

A novel polymer-based probe for fluorescently ratiometric sensing of hydrogen sulfide with multiple applications

Summary: A novel polymer-based probe P1 was successfully synthesized for fluorescently ratiometric sensing of H2S with high selectivity and sensitivity. A smartphone sensing platform was constructed to conduct visual quantitative detection of H2S. P1 can be employed in evaluating the level fluctuations of H2S in living cells, testing water samples/wine samples, and monitoring food freshness.

Split aptamer-based sandwich-type ratiometric biosensor for dual-modal photoelectrochemical and electrochemical detection of 17β-estradiol

Summary: The study developed a split aptamer-based sandwich-type ratiometric biosensor for the detection of 17 beta-estradiol (E2) using photoelectrochemical and electrochemical assays. The biosensor utilized split aptamer fragments to recognize E2 and trigger a hybridization chain reaction, resulting in the production of double-stranded DNA labeled with CdTe quantum dots. This DNA complex was able to sensitize CdTe quantum dots and generate response signals for E2 detection. The developed biosensor demonstrated high sensitivity and accuracy with two linear ranges and low detection limits.
