Summary: In this study, the expression profiles of miRNAs in serum samples of bladder cancer patients and healthy controls were detected, and five differentially expressed miRNAs were identified. A three-miRNA signature consisting of miR-27b-3p, miR-381-3p, and miR-451a was constructed with a high diagnostic efficiency for bladder cancer. This three-miRNA signature may serve as a promising non-invasive biomarker for early diagnosis of bladder cancer.

Summary: This study found that the expression levels of miR-128-3p and miR-33a-5p in serum exosomes were significantly different between non-small cell lung cancer (NSCLC) patients and healthy volunteers, suggesting their potential as biomarkers for NSCLC. The combination of miR-128-3p and miR-33a-5p detection improved the accuracy of NSCLC screening. These findings provide new insights for the early diagnosis and treatment of NSCLC.

This review presents recent strategies and advances in plasmonic nanoparticle-based miRNA delivery for cancer detection and treatment, and discusses the challenges and potential opportunities in the field of RNAi-based theragnosis.

Summary: This study developed a high-throughput drug screening method using chordoma models to identify new combination therapies for chordoma. Several combination therapies, including PI3K inhibitors with other drugs, were identified as having potential therapeutic efficacy. ALK, PDGFR, VEGFR, aurora kinase, and BCL-2 were identified as important targets. T (brachyury) inhibition reduced cell viability, but did not show additional benefit with PI3K pathway inhibition in this model.

Summary: AK-2292 is a potent and selective small-molecule degrader of both STAT5A and STAT5B isoforms. It selectively inhibits STAT5 activity and induces degradation of STAT5A/B proteins without affecting other STAT proteins and numerous non-STAT proteins. AK-2292 effectively induces STAT5 depletion in both normal tissues and CML xenograft tissues, leading to tumor regression in CML xenograft mouse models. It is not only a powerful research tool to study STAT5 biology and therapeutic potential, but also a promising lead compound for STAT5-targeted therapy development.

Downregulation of NEDD8 suppresses tumor growth and induces cell cycle arrest, DNA damage, and apoptosis in ESCC. The mechanism involves the accumulation of CRLs substrates, which can be attenuated by knockdown of DR5.
