In recent years, advanced radiation therapy techniques, including stereotactic body radiotherapy and carbon-ion radiotherapy, have progressed to such an extent that certain types of cancer can be treated with radiotherapy alone. The therapeutic outcomes are particularly promising for early stage lung cancer, with results matching those of surgical resection. Nevertheless, patients may still experience local tumor recurrence, which might be exacerbated by the acquisition of radioresistance after primary radiotherapy. Notwithstanding the risk of tumors acquiring radioresistance, secondary radiotherapy is increasingly used to treat recurrent tumors. In this context, it appears essential to comprehend the radiobiological effects of repeated photon and particle irradiation and their underlying cellular and molecular mechanisms in order to achieve the most favorable therapeutic outcome. However, to date, the mechanisms of acquisition of radioresistance in cancer cells have mainly been studied after repeated in vitro X-ray irradiation. By contrast, other critical aspects of radioresistance remain mostly unexplored, including the response to carbon-ion irradiation of X-ray radioresistant cancer cells, the mechanisms of acquisition of carbon-ion resistance, and the consequences of repeated in vivo X-ray or carbon-ion irradiation. In this review, we discuss the underlying mechanisms of acquisition of X-ray and carbon-ion resistance in cancer cells, as well as the phenotypic differences between X-ray and carbon-ion-resistant cancer cells, the biological implications of repeated in vivo X-ray or carbon-ion irradiation, and the main open questions in the field.

Summary: A combination of high-energy carbon ion radiation therapy and checkpoint inhibitors shows significant effectiveness in reducing the growth of abscopal tumors in a murine model, with carbon ions being more efficient in reducing lung metastases compared to x-rays. The infiltration of immune cells in the abscopal tumors increased in animals treated with the combination therapy, suggesting a potential for effective treatment of advanced tumors with this approach.

Summary: A selenium deficient mouse model was successfully created by starting the mother on a selenium deficient diet 2 weeks after giving birth, showing sensitivity to oxidative stress induced by H2O2 and prolonged survival. This model may be useful for studying oxidative stress in vivo.

Summary: The study used a small-molecule radiotracer, C-11-l-1MTrp, to quantitatively and longitudinally visualize whole-body IDO1 dynamics, accurately delineating changes in the cancer-immune milieu and assessing the efficacy of multiple combinatorial immunotherapies. The results suggest that PET imaging of IDO1 with C-11-l-1MTrp can serve as a robust method for tracking IDO1 in therapeutic interventions, highlighting its potential as a new biomarker for personalized cancer immunotherapy strategies.

Effect of Three Types of Ion Beam Irradiation on Gerbera (Gerbera hybrida) In Vitro Shoots with Mutagenesis Efficiency

Summary: In this study, Gerbera in vitro shoots were irradiated with ion beams of different LETs to explore effective LET and absorbed doses for mutagenesis. It was found that ion beams with higher LETs had a lower LD50, and induced trait/morphological mutations in color, shape, and male fertility. The highest growth change and trait/morphological mutation rates were observed with C irradiation at 10 Gy, Ar irradiation at 5 Gy, and Fe irradiation at 5 Gy, with Ar irradiation at 5 Gy showing the highest mutation rate of 14.1%. Genomic mutations in flower color, petal shape, and male sterile mutants were confirmed using Genotyping by Random Amplicon Sequencing-Direct technology. These findings could lead to more efficient gerbera mutant production and analysis technology.
