Summary: The study identified a novel lncRNA, lncROPM, highly expressed in breast cancer stem cells (BCSCs), which regulates PLA2G16 expression to promote phospholipid metabolism and activate multiple signaling pathways involved in maintaining BCSC properties. Targeting the lncROPM-PLA2G16 signaling axis may be a novel therapeutic strategy for breast cancer patients.

Summary: This study explored the relationship between angiogenesis-related lncRNAs (AR-lncRNAs) and breast cancer (BC) immunity and prognosis. A total of 17 AR-lncRNAs closely associated with BC prognosis were identified. BC patients were divided into five clusters based on the expression of these prognostically relevant AR-lncRNAs. An AR-lncRNA risk model (AR-lncM) was created using LASSO regression analysis and validated for its predictive performance. The gene LINC01614 was found to regulate epithelial-mesenchymal transition (EMT) and tamoxifen sensitivity in BC cells, suggesting its potential as a therapeutic target.

Summary: HOTAIR is an oncogenic non-coding RNA that is strongly associated with tumor grade and prognosis in various carcinomas, including breast cancer (BC). It regulates target genes through sponging and epigenetic mechanisms and controls oncogenic cellular and signaling mechanisms, such as metastasis and drug resistance. In BC cells, HOTAIR expression is regulated by transcriptional and epigenetic mechanisms. This review discusses the regulatory mechanisms of HOTAIR during cancer development and explores its role in BC development, metastasis, and drug resistance. The review also highlights the potential therapeutic applications of HOTAIR in BC management, treatment, and prognosis.

Summary: Among TNBC subtypes, the BL2 subtype has the lowest survival rate and highest risk of metastasis after chemotherapy treatment. Alpha B-crystallin (CRYAB) is highly expressed in BL2 subtypes and is associated with brain metastasis in TNBC patients. We hypothesized that alpha B-crystallin is associated with increased cell motility in the BL2 subtype after chemotherapy.

Summary: This study developed a new therapy using a combination of verteporfin and CA4 in polymer-lipid hybrid nanoparticles, which effectively inhibited TNBC cell viability and migration, and decreased paclitaxel-induced and/or CA4-induced CSC enrichment. The approach not only targeted bulk cancer cells, CSCs, and angiogenesis, but also showed potential in clinically relevant patient-derived xenograft models.

Summary: Mitochondrial dysfunction can transform cancer cells into cancer stem-like cells, as shown by the generation of mitochondria dysfunctional (rho(0)) cells from the Hep3B hepatocellular carcinoma cell line. These rho(0) cells exhibited increased cancer stem-like properties, such as self-renewal, chemotherapy resistance, and angiogenesis. The data suggest that mitochondrial dysfunction plays a significant role in driving the transition of cancer cells towards a stem-like phenotype.

Summary: Ovarian cancer has a high mortality rate and lacks effective targeted therapy. This study aimed to explore genetic markers and their clinical value in ovarian cancer development. 11 genetic markers, including ACTL6A, were found to be significantly elevated in invasive and chemoresistant ovarian cancer. ACTL6A expression was correlated with survival prognosis, and HER2 inhibitor CP724714 showed potential therapeutic benefits in high ACTL6A expression cases. High ACTL6A expression can serve as a biomarker for predicting treatment-refractory, metastasis, and prognosis of ovarian cancer.
