Functionalized manganese iron oxide nanoparticles: a dual potential magneto-chemotherapeutic cargo in a 3D breast cancer model

Summary: This study investigated the effectiveness of MIONP-based magneto-chemotherapy (MCT) in a 3D breast cancer model, using advanced 3D in vitro tumor models as a promising alternative to replicate the complete range of tumor characteristics. The results demonstrated that MCT using MIONPs reduced the viability of breast cancer cells, with the most promising therapeutic effects observed in the 3D cancer model.

Summary: Localized heat generation from manganese iron oxide nanoparticles conjugated with chemotherapeutics under the exposure of an alternating magnetic field can revolutionize targeted breast cancer therapy. Advanced 3D in vitro tumor models are explored as a promising alternative to replicate the complete range of tumor characteristics. The effectiveness of magneto-chemotherapy using chitosan-coated MIONPs is investigated in a 3D breast cancer model, with successful reduction in cancer cell viability.

Unlocking Synergistic Potential: Agomelatine Enhances the Chemotherapeutic Effect of Paclitaxel in Breast Cancer Cell Through MT1 Melatonin Receptors and ER-alpha Axis

Summary: This study investigates the potential of combining agomelatine (AGO), a synthetic melatoninergic drug, with paclitaxel (PTX) for the treatment of breast cancer. The results show that AGO has cytotoxic effects on breast cancer cells similar to melatonin (MTN), and combining AGO and MTN with PTX shows synergistic effects. The study also reveals differences in molecular mechanisms and the importance of MTN receptors in breast cancer cell types.

Simulation of drug-drug interactions between breast cancer chemotherapeutic agents and antiemetic drugs

Summary: This study evaluated the potential drug-drug interactions between breast cancer chemotherapy drugs and antiemetic agents. The results showed that about half of the drugs had no potential for DDI, while around 30% of the drugs were not metabolized by CYPs. However, some antiemetic drugs had moderate to no interactions with anticancer agents. Therefore, clinicians need to be aware of possible DDI when using drug combinations for breast cancer treatment.

Summary: Acquired resistance to PAL or ABE in breast cancer cells confers cross-resistance to the other CDK4/6 inhibitor but not to chemotherapeutic agents. Down-regulation of basal RB expression and normalized RB phosphorylation reduced by CDK4/6 inhibitors may be responsible for the attenuated anti-cell growth effects of the inhibitors.

These salts can be bioactivated by CYP1A1 into potent antitumor metabolites. The PAIB-SA salts have the potential to be developed as suitable formulations for further biopharmaceutical and pharmacodynamic studies.
