Summary: Juvenile myelomonocytic leukemia (JMML) is a rare childhood neoplasm categorized under myelodysplastic/myeloproliferative neoplasms. A low-cost targeted myeloid panel was developed to comprehensively evaluate the genomic profile of JMML, revealing that almost 90% of patients have at least one mutation, with the most common mutations found in RAS/MAPK-pathway genes. Monosomy 7 was detected in 32% of patients, with implications for overall survival.

Summary: MicroRNAs, small non-coding RNA molecules, are increasingly used as biomarkers in diseases with challenges due to their low abundance in serum/plasma. This study compared four commercial kits and found that the miRNeasy Serum/Plasma kit performed the best in terms of quality and quantity of microRNA isolated, with a modified protocol further enhancing its performance.

The study found that the kappa B.1.1671.1 and delta B.1.617.2 variants were prevalent among both vaccinated and unvaccinated individuals. The second dose of the vaccine was associated with a lower severity of COVID-19.

Summary: Multicolor flow cytometry-based DNA-ploidy (MFC-ploidy) analysis shows a characteristic pattern in samples with endoreduplication and can distinguish between masked hypodiploidy and true hyperdiploidy in B-cell acute lymphoblastic leukemia (B-ALL). An integrated approach involving cytogenetic and MFC-ploidy detection is highly valuable for risk stratification of B-ALL in routine clinical practice.

Summary: In this study, the long-term toxicities of imatinib treatment in children with pCML were evaluated. The results showed that a significant proportion of patients did not achieve deep molecular response, and some patients had to switch to second-generation TKIs due to poor response. Low bone density and hypovitaminosis D were common long-term toxicities.

Summary: Exon 17 KIT mutations play an important role in predicting relapse in pediatric AML with RUNX1::RUNX1T1 fusion, and a high KIT variant-allele frequency may indicate poor prognosis in these patients. These findings emphasize the need to incorporate KIT mutational analysis into risk stratification for pediatric CBF-AML.
