Microbe-MUC1 Crosstalk in Cancer-Associated Infections

Infection-associated cancers account for similar to 20% of all malignancies. Understanding the molecular mechanisms underlying infection-associated malignancies may help in developing diagnostic bio-markers and preventative vaccines against malignancy. During infection, invading microbes interact with host mucins lining the glandular epithelial cells and trigger inflammation. MUC1 is a transmembrane mucin glycoprotein that is present on the surface of almost all epithelial cells, and is known to interact with invading microbes. This interaction can trigger pro- or anti-inflammatory responses depending on the microbe and the cell type. In this review we summarize the mechanisms of microbe and MUC1 interactions, and highlight how MUC1 plays contrasting roles in different cells. We also share perspectives on future research that may support clinical advances in infection-associated cancers.

Repression of MUC1 Promotes Expansion and Suppressive Function of Myeloid-Derived Suppressor Cells in Pancreatic and Breast Cancer Murine Models

Summary: The study revealed accelerated tumor growth in MUC1KO mice, associated with increased suppressive MDSCs and Tregs in the tumor microenvironment. MUC1KO host showed elevated levels of iNOS, ARG1, and TGF-beta, promoting proliferation of MDSCs with an immature and immune suppressive phenotype. Additionally, MDSCs from MUC1KO mice exhibited higher c-Myc and activated pSTAT3 levels, suggesting increased survival, proliferation, and prevention of maturation of MDSCs in the MUC1KO host.

Overexpression of MUC1 Induces Non-Canonical TGF-β Signaling in Pancreatic Ductal Adenocarcinoma

Summary: This study finds that MUC1 plays a significant role in the development of pancreatic ductal adenocarcinoma (PDA). Overexpression of MUC1 activates the JNK pathway and increases cell viability. Conversely, low expression of MUC1 allows TGF-beta to retain its tumor-suppressive function. These findings contribute to a better understanding of the mechanisms behind PDA development.

Targeting tumor-associated MUC1 overcomes anoikis-resistance in pancreatic cancer

Summary: Pancreatic cancer, particularly pancreatic ductal adenocarcinoma (PDA), is a major cause of cancer-related deaths in the United States. Surgical resection is the best treatment option, but only a small percentage of patients are eligible. Recurrence and metastasis are the main reasons for mortality. A monoclonal antibody named TAB004 has been developed to target tumor-associated MUC1 (tMUC1), which is expressed in over 80% of PDAs. Treatment with TAB004 has shown significant effects in reducing colony-forming potential in PDA cells and inducing cell death. The mechanism involves disruption of cellular integrity, activation of ER stress and anoikis, and inhibition of oncogenic signaling pathways. TAB004 also exhibited promising results in mouse models, indicating its potential as a prophylactic agent to prevent tumor relapse, metastasis, and increase the efficacy of chemotherapy.
