Extraction, Characterization, Antitumor and Immunological Activities of Hemicellulose Polysaccharide from Astragalus radix Herb Residue

Astragalus radix (radix) have been frequently used for clinical application in China, and the herb residues of radix turn out to be a waste of resources. To escape from this, the medicine value of radix herb residues is mined in this article. We isolated hemicellulose polysaccharide AX-I-3b from radix herb residues by fractional extraction. Monosaccharide-composition analysis revealed that AX-I-3b consisted of arabinose, xylose, and glucose with a molar ratio of 10.4:79.3:1.1. Methylation, NMR and FT-IR analyses showed that AX-I-3b monosaccharide residue was linked as follows: -> 2,3,4)-beta -d-Xylp-(1 ->, -> 4)-beta -d-Arap-(1 ->, -> 4)-beta -d-Glcp-(1 ->. Then, we found that AX-I-3b exhibited antitumor activity against lung cancer in vitro and vivo through MTT assay and xenograft tumor model. Mechanistically, AX-I-3b induced apoptosis in lung cancer cells and xenograft tumors, which is evidenced by the up-regulation of p53, Bax and cleaved caspase-3, and the down-regulation of Bcl-2. Moreover, AX-I-3b synergistically improved the therapeutic ability of cisplatin in xenograft tumors model. Furthermore, AX-I-3b treatment effectively improved the immune organ index, the percentage of spleen lymphocyte subsets and serum cytokine levels in lung cancer mice, supporting that AX-I-3b showed immunomodulatory activity. In conclusion, our results identified AX-I-3b as an antitumor and immunomodulatory agent, providing a new insight into the reutilization of radix herb residue.

Deciphering the correlations between aging and constipation by metabolomics and network pharmacology

Structure-activity relationship of novel acridone derivatives as antiproliferative agents

Combination of Compound Kushen Injection and cisplatin shows synergistic antitumor activity in p53-R273H/P309S mutant colorectal cancer cells through inducing apoptosis

Summary: This study aimed to investigate the combined efficacy of Compound Kushen Injection (CKI)-derived combination strategies in p53-mutant CRC. The results showed that CKI combined with cisplatin (Cis) produced synergistic antitumor efficiency for p53-R273H/P309S mutant CRC cells. The combination of CKI and Cis reinforced cell death in a synergistic manner by inducing extrinsic apoptosis pathway.

Comprehensive 16S rRNA sequencing based microbiomes and 1H NMR based metabolomics reveal the relationships of aging and constipation

Summary: This study found a close relationship between gut microbiota and aging as well as constipation, possibly through metabolism. The composition of gut microbiome was similar in both aging and constipation, and key gut bacteria were clustered together. Gut microbial metabolites played important roles as signaling molecules in aging and constipation, and the occurrence and development of both conditions were influenced by multiple factors.

Microbiome and metabolome reveal the metabolic and microbial variations induced by depression and constipation

Summary: This study aims to reveal the possible correlations between depression and constipation from the perspectives of gut microbiome and plasma metabolome. Through constructing depressed and constipated rat models, locomotor activity and gastrointestinal functions were measured. Plasma metabolomics and gut microbiota sequencing were used to identify shared and unique metabolites between depression and constipation. The findings show that depression and constipation significantly affect locomotor activity, gastrointestinal functions, and gut microbiota compositions of rats, with perturbations in energy metabolism and amino acid metabolism.

Discovering the Mechanisms of Oleodaphnone as a Potential HIV Latency-Reversing Agent by Transcriptome Profiling

Gut microbiome and tissue metabolomics reveal the compatibility effects of Xiaoyaosan on depression based on ?gut-liver-kidney? axis

Summary: This study aimed to reveal the compatibility of Xiaoyaosan (XYS) in anti-depression through the perspective of the gut-liver-kidney axis. Results showed that XYS and its efficacy groups significantly regulated the abnormalities of behaviors, kidney metabolism, and intestinal disorders in depressed rats. XYS exhibited the strongest regulation, followed by the Shugan (SG) and Jianpi (JP) groups. The findings provide innovative insights into the anti-depression effects and compatibility of XYS, as well as a new perspective and methodology for studying complex diseases and traditional Chinese medicine (TCM) compatibility.

Microbiome-metabolomics deciphers the effects of Cistanche deserticola polysaccharides on agedconstipated rats

Summary: This study investigated the impact of constipation in an aged rat model and the effects of Cistanche deserticola polysaccharide (CDPS) on constipation. The study found that CDPS significantly regulated behavioral indexes, gut microbiota, and fecal metabolites in constipated rats. It increased beneficial bacteria and decreased potentially pathogenic bacteria. 16 potential biomarkers of constipation in aged rats were identified. The findings deepen our understanding of constipation in the elderly and provide a foundation for using polysaccharides to treat constipation and improve the quality of life of the elderly.
