Early Stage Adaptation of a Mesophilic Green Alga to Antarctica: Systematic Increases in Abundance of Enzymes and LEA Proteins

It is known that adaptive evolution in permanently cold environments drives cold adaptation in enzymes. However, how the relatively high enzyme activities were achieved in cold environments prior to cold adaptation of enzymes is unclear. Here we report that an Antarctic strain of Chlorella vulgaris, called NJ-7, acquired the capability to grow at near 0 degrees C temperatures and greatly enhanced freezing tolerance after systematic increases in abundance of enzymes/proteins and positive selection of certain genes. Having diverged from the temperate strain UTEX259 of the same species 2.5 (1.1-4.1) to 2.6 (1.0-4.5) Ma, NJ-7 retained the basic mesophilic characteristics and genome structures. Nitrate reductases in the two strains are highly similar in amino acid sequence and optimal temperature, but the NJ-7 one showed significantly higher abundance and activity. Quantitative proteomic analyses indicated that several cryoprotective proteins (LEA), many enzymes involved in carbon metabolism and a large number of other enzymes/proteins, were more abundant in NJ-7 than in UTEX259. Like nitrate reductase, most of these enzymes were not upregulated in response to cold stress. Thus, compensation of low specific activities by increased enzyme abundance appears to be an important strategy for early stage cold adaptation to Antarctica, but such enzymes are mostly not involved in cold acclimation upon transfer from favorable temperatures to near 0 degrees C temperatures.

Summary: For Enterococcus faecalis, sodium hypochlorite, hydrogen peroxide, and chlorhexidine gluconate have different minimum inhibitory concentrations, but at their respective minimum inhibitory concentrations, the bacteria may still be in a non-growing but metabolically active state, which can lead to recurring infections. In contrast, at their respective minimum inhibitory concentration based on metabolic activity, all three disinfectants completely inhibit the metabolic activity of the bacteria. The combination of these disinfectants at lower concentrations can achieve an equivalent metabolism-inhibiting effect as using each disinfectant alone at the minimum inhibitory concentration, thereby controlling the infection of E. faecalis and reducing side effects.

Assessing Efficacy of Clinical Disinfectants for Pathogenic Fungi by Single-Cell Raman Microspectroscopy

Experimental results with clinical RNA samples showed that the instrument can effectively diagnose COVID-19 patients.

Medium-chain triglyceride production in Nannochloropsis via a fatty acid chain length discriminating mechanism

Summary: Depending on their chain length, triacylglycerols (TAGs) have different applications, so it is desirable to have a genetically designed feedstock with specific chain length for maximizing process efficiency and product versatility. In the microalga Nannochloropsis oceanica, two enzymes were found to assemble medium-chain fatty acids (MCFAs) into TAGs, with one being a generalist and the other a specialist. By manipulating these enzymes, the production of medium-chain triacylglycerols (MCTs) in TAGs can be significantly increased in microalgae, allowing for the production of designer oils.

Auto Flow-Focusing Droplet Reinjection Chip-Based Integrated Portable Droplet System (iPODs)

Summary: Droplet microfluidics is a powerful tool for biochemical applications, but precise fluid control is a hindrance in droplet-based point-of-care testing. This paper presents a droplet reinjection method that allows for droplet alignment and detection without precise fluid control or external pumps. By integrating a surface-wetting-based droplet generation chip, an integrated Portable Droplet system (iPODs) is developed, which can generate monodisperse droplets with a narrow size distribution and high stability. The iPODs also demonstrates good linearity in the digital loop-mediated isothermal amplification process, highlighting its potential as a portable, low-cost, and easy-to-deploy toolbox for droplet-based applications.
