Summary: In this study, comprehensive pan-cancer analyses were conducted to examine the expression and prognostic implications of OLR1. The results showed that OLR1 is highly expressed in most cancers and is associated with patient prognosis. OLR1 expression is positively correlated with immunosuppressive cell infiltration and immune checkpoint molecules, while being negatively associated with effector T cells. Moreover, OLR1 expression is also correlated with tumor mutation burden and microsatellite instability in some cancers, and it affects EMT, stemness, and cuproptosis resistance outcomes.

Summary: The UBQLN family of proteins, including UBQLN1 and UBQLN2, has important regulatory roles in cancer progression and tumorigenesis. Loss of UBQLN1/UBQLN2 leads to increased cell viability, proliferation, migration, clonogenic potential, and cell cycle progression, which is associated with increased MYC expression. UBQLN1 and UBQLN2 interact with phosphorylated MYC and facilitate its degradation. Overexpression of UBQLN1 reverses the increased expression of MYC following the loss of UBQLN2. Loss of UBQLN1 drives tumorigenesis and lung metastasis in mice, which is associated with increased expression of MYC, proteins involved in cell cycle progression, and EMT. These findings suggest a novel role of UBQLN1 and UBQLN2 in regulating MYC in lung adenocarcinoma cells.

Summary: This study reveals the metastasis-promoting role of circMGEA5 in osteosarcoma. CircMGEA5 acts as a competing endogenous RNA by directly sponging miR-153-3p and miR-8084, leading to the upregulation of ZEB1 and Snail, thus inducing epithelial-mesenchymal transition and metastasis. The depletion of circMGEA5 significantly reduces lung metastasis in osteosarcoma cells. Targeting this newly identified ceRNA axis may be crucial in the development of novel therapies for metastatic osteosarcoma patients.
