Summary: This study aims to explore the diagnostic and prognostic value of plasma miRNA in colorectal cancer (CRC) patients and establish a prognostic model. The results showed that miR-96/miR-99b can be used as a promising biomarker for early detection of metastatic CRC (mCRC) patients, and the nomogram based on 5 characteristics has satisfactory prognostic value.

Summary: This study aimed to determine the relative quantitative expression levels of three miRNAs (miR-601, 760, and 106b-5p) in the serum of NSCLC patients and compare them to controls. The results showed downregulation of miR-601 and miR-760 and upregulation of miR-106b-5p in NSCLC patients. MiR-760 exhibited the highest diagnostic and prognostic potential.

Summary: This study identified a miR-29a-PTEN-AKT axis as a downstream signaling pathway of ER alpha in breast cancer metastasis. Low level of miR-29a was associated with reduced metastasis and better survival in ER alpha+ luminal subtype of BC, while high level of miR-29a was detected in ER alpha- triple negative breast cancer (TNBC) with distant metastasis and poor survival. Targeting miR-29a suppressed cellular invasion, EMT and lung metastasis in TNBC cells in vitro and in TNBC tumor-bearing mice in vivo.

Summary: HOTAIR is an oncogenic non-coding RNA that is strongly associated with tumor grade and prognosis in various carcinomas, including breast cancer (BC). It regulates target genes through sponging and epigenetic mechanisms and controls oncogenic cellular and signaling mechanisms, such as metastasis and drug resistance. In BC cells, HOTAIR expression is regulated by transcriptional and epigenetic mechanisms. This review discusses the regulatory mechanisms of HOTAIR during cancer development and explores its role in BC development, metastasis, and drug resistance. The review also highlights the potential therapeutic applications of HOTAIR in BC management, treatment, and prognosis.

Summary: The downregulation of miR-601 in osteosarcoma was found in this study. Patients with lower expression of miR-601 had worse prognosis. In vitro and in vivo experiments showed that elevated miR-601 inhibited the proliferative, migratory, and invasive capacities of osteosarcoma cells. These effects were mediated by miR-601 targeting PKMYT1.

Summary: This study aimed to identify differentially expressed genes and ncRNAs in breast cancer tissues compared to normal adjacent tissues, and found miR-4510 as a potential biomarker for breast cancer. The study used MACE-Seq and small RNA-Seq to analyze gene expression and identified miR-4510 as significantly down-regulated in breast cancer. Experimental data suggested that miR-4510 may function as a tumor suppressor in breast cancer.

Summary: The study revealed that circRNA circFOXK2 plays a crucial role in BC metastasis, with its upregulation associated with poor clinicopathological characteristics. Functional experiments demonstrated that overexpression of circFOXK2 could promote migration and invasion of BC cells, and its interaction with IGF2BP3 and miR-370 synergistically enhances BC metastasis. These findings suggest circFOXK2 as a potential biomarker for the diagnosis and treatment of BC.

Summary: Breast cancer is the leading cause of death by cancer in women globally. A miRNA signature deregulated in preclinical and clinical BCa models was identified for potential biomarker discovery, useful for BCa diagnosis and/or prognosis. Certain miRNAs were up-regulated and down-regulated in BCa, showing negative correlation with the expression of related genes. Genes such as GAB1, GNG12, HBP1, and SESN1 have been found to have expression levels related to patient survival in aggressive BCa subtypes.

Summary: Myo-Inositol inhibits the PI3K/Akt pathway, particularly effective in triple-negative breast cancer cells, but not in hormone-responsive breast cancer cells unless estrogen effects are counteracted. This novel axis involving miR-125a-5p downregulating IP(6)K1 gene could be activated as a molecular target therapy in breast cancer, inhibiting metastasis and tumor progression.

Summary: A new nomogram prognostic model for breast cancer patients with brain metastasis was developed and validated, showing good performance in risk stratification and treatment strategies. The model included various variables such as age, race, surgery, and metastatic sites, with a C-index of 0.69 in the development cohort and 0.70 in the validation cohort.

Summary: The study identified high levels of miR-211 as a prognostic marker for breast cancer brain metastasis, enhancing trans-blood-brain barrier migration and adherence of tumor cells leading to brain colonization and poor survival outcomes. Furthermore, downstream targets of miR-211, SOX11 and NGN2, play a role in regulating TNBC brain metastasis.

Summary: Growing evidence suggests that LINC00659, a newly identified long noncoding RNA, plays a critical role in gastric cancer. Upregulation of LINC00659 is associated with tumor progression, lymphatic metastasis, and poorer prognosis. It functions as a competing endogenous RNA for miR-370, leading to the upregulation of its target gene AQP3. This study provides insights into the pathogenesis of gastric cancer and may contribute to the identification of potential therapeutic targets.

Summary: Ischemic stroke is a common and serious disease, and neuroinflammation plays a crucial role in its progression. Microglia, astrocytes, and infiltrating immune cells are involved in the complicated neuroinflammation cascade, releasing different molecules that affect inflammation. Flavonoids, plant-specific compounds, have shown protective effects against cerebral ischemia injury by modulating the inflammatory responses.
