For psychiatric disorders, repeated failures in converting scientific discoveries into novel drugs has precipitated a crisis and eroded confidence in drug discovery. This review describes how current and future innovations driven by application of biomarkers can help to re-initiate research in this area. This will have positive impact on the field of psychiatry and result in application of sensitive and specific biochemical tests in parallel with the traditional questionnaires for improved diagnosis. Furthermore, application of emerging biosensor tools will facilitate point-of-care testing by fusion of biochemical and clinical data. In this way, patient data will be comprised of past medical histories, biopatterns and prognosis information, resulting in personalized profiles or molecular fingerprints for patients with these conditions.

Induced pluripotent stem cell-derived astrocytes from patients with schizophrenia exhibit an inflammatory phenotype that affects vascularization

Summary: This study examined the proteome, inflammatory responses, and secretome effects of astrocytes derived from schizophrenia patients. The findings suggest that these astrocytes exhibit molecular and functional abnormalities, affecting immune function and vascularization. Reduced expression of NF-kappa B p65 subunit was observed, and the secretion of interleukin 8 (IL-8) was particularly elevated. The conditioned medium from these astrocytes could reduce the diameter of newly grown vessels.

Mori

Summary: Visceral adiposity is a risk factor for severe COVID-19, and the degree of adipose tissue infection and cellular response to SARS-CoV-2 depend on the adipose tissue depot and viral lineage. Visceral fat cells are more susceptible to SARS-CoV-2 infection and show higher expression of pro-inflammatory cytokines. Cell death occurs to a similar degree regardless of cell origin or viral lineage.

Protein Succinylation and Malonylation as Potential Biomarkers in Schizophrenia

Bradley Joseph Smith, Caroline Brandao-Teles, Giuliana S. Zuccoli, Guilherme Reis-de-Oliveira, Mariana Fioramonte, Veronica M. Saia-Cereda, Daniel Martins-de-Souza

Summary: Lysine succinylation and malonylation, two protein post-translational modifications, play important roles in protein regulation, glycolysis, and energy metabolism. The precursors of these modifications, succinyl-CoA and malonyl-CoA, are key players in central metabolic processes and are responsive to metabolic stimuli. As mitochondrial dysfunction and metabolic dysregulation are involved in psychiatric illnesses like schizophrenia, these modifications can provide insights into protein regulation and serve as potential biomarkers for disease diagnosis, progression, and treatment. The study analyzed data from shotgun mass spectrometry-based quantitative proteomics to investigate the succinylome and malonylome of postmortem brain tissue from schizophrenia patients, controls, and a schizophrenia chemical model, as well as the human oligodendrocyte precursor cell line MO3.13 with antipsychotics and co-treatments. The results revealed significant changes in the succinylome and malonylome, highlighting the importance of these understudied protein modifications.

Proteomic signatures of schizophrenia-sourced iPSC-derived neural cells and brain organoids are similar to patients' postmortem brains

Juliana Minardi Nascimento, Veronica M. Saia-Cereda, Giuliana S. Zuccoli, Guilherme Reis-de-Oliveira, Victor Corasolla Carregari, Bradley J. Smith, Stevens K. Rehen, Daniel Martins-de-Souza

Summary: This study evaluates the differential protein expression in neural progenitors, early neurons, and brain organoids derived from schizophrenia patients, offering insights into the developmental origins of the disorder. The results indicate dysregulated proteins and impaired pathways related to spliceosomes, amino acid metabolism, axonal guidance, and synaptogenesis. The comprehensive protein-level data provided by this study contributes to understanding the early events in brain development and the mechanisms underlying schizophrenia.

Mitochondrial, cell cycle control and neuritogenesis alterations in an iPSC-based neurodevelopmental model for schizophrenia

Giuliana S. S. Zuccoli, Juliana M. M. Nascimento, Pedro M. M. Moraes-Vieira, Stevens K. K. Rehen, Daniel Martins-de-Souza

Summary: Schizophrenia is a severe neurodevelopmental psychiatric disorder affecting around 1% of the global population. Proteomic studies on neural stem cells and neurons derived from induced pluripotent stem cells from schizophrenia patients have revealed differentially expressed proteins related to mitochondrial function, cell cycle control, DNA repair, and neuritogenesis. Functional analysis of neural stem cells also showed alterations in mitochondrial oxygen consumption and higher levels of intracellular reactive oxygen species in schizophrenia-derived cells.

Summary: Research has found that SARS-CoV-2 mainly infects T helper cells (CD4+), but not T cells (CD8+). The infected T helper cells express higher levels of IL-10, which may contribute to viral persistence and increased disease severity, leading to poor immune response in COVID-19 patients.
