Effects of the Hyptis martiusii Benth. leaf essential oil and 1,8-cineole (eucalyptol) on the central nervous system of mice

The aim of this study was to characterize the central effects of the Hyptis martiusii leaf essential oil (OEHM) and 1,8-cineole (eucalyptol) using behavioral animal models. Gas chromatography coupled to mass spectrometry (GC/MS) was used to characterize the chemical compounds present in the OEHM. For the behavioral tests, female Swiss mice treated with the OEHM (25, 50, 100 and 200 mg/kg, i.p.) and 1,8-cineole (50 mg/kg, i.p.) were used and subjected to the following tests: open field, elevated cross maze, rotarod, sodium pentobarbital- or ethyl ether-induced sleep time, pentylenetetrazol-induced convulsions, haloperidol-induced catalepsy, and ketamine-induced hyperkinesia. GC/MS analysis identified 20 constituents with the majority of them being monoterpenes and sesquiterpenes, with eucalyptol (1,8-cineol), the major sample compound (25.93%), standing out. The results showed the OEHM (25, 50 100 and 200 mg/kg, i.p.) and its major compound (50 mg/kg, i.p.) reduced animal motility in the open field test, increased pentobarbital- and ethyl ether-induced sleep time, as well as death latency in the pentylenetetrazole-induced convulsion model. However, the tested compounds were devoid of anxiolytic-like and myorelaxant activity. In addition, the OEHM (100 and 200 mg/kg, i.p.) and 1,8-cineole (50 mg/kg, i.p.) potentiated haloperidol-induced catalepsy and reduced ketamine-induced hyperkinesia. Taken together, the results suggest the OEHM has important hypnotic-sedative and antipsychotic-like effects, which appear to be due to the monoterpene 1,8-cineole, the major compound identified in the essential oil.

Chemical composition and biological activities of the essential oil from Eugenia stipitata McVaugh leaves

Summary: This study investigated the volatile components and activities of the essential oil obtained from the leaves of Eugenia stipitata McVaugh. The oil showed high selective activity against Leishmania braziliensis and good antibacterial activity. However, its free-radical scavenging activity was low. These findings suggest that the essential oil of Eugenia stipitata is a valuable source for the development of antibacterial and antiprotozoal drugs.

The conversion of cryogels into affinity adsorbents, particularly using epoxy radicals as supports, provides a promising alternative for biomolecule purification.

Summary: Alzheimer's and Parkinson's are neurodegenerative disorders that seriously affect people's quality of life. This review used Molecular Docking to evaluate the potential of linalool, citronellal, and their derivatives as anti-Alzheimer's and anti-Parkinson's agents. The compounds demonstrated good pharmacokinetic characteristics and showed strong affinity for targets involved in the pathophysiology of these diseases. Linalool and its derivatives also exhibited promise against the BACE enzyme activity associated with Alzheimer's disease. These compounds have the potential to be developed into future drugs for the treatment of neurodegenerative disorders.

Summary: The increase in antibiotic resistance rates has led researchers to search for antibacterial compounds that can enhance the activity of conventional antibiotics. Coumarin derivatives have been found to be effective antibacterials with potential new mechanisms of action for treating drug-resistant bacterial infections. This study synthesized new coumarin derivatives and evaluated their pharmacokinetic properties, chemical similarity, antimicrobial activity, and potential for modulating antibiotic resistance using in vitro assays.

Summary: The discovery of antibiotics has greatly improved the treatment of bacterial infections, but the emergence of antibiotic resistance has reduced their effectiveness. This study investigated the antibacterial activity of synthetic coumarins against Staphylococcus aureus and their interaction with the MepA efflux pump. The results showed that the coumarin derivatives did not have direct antibacterial activity, but they enhanced the activity of an efflux inhibitor and potentiated the activity of ciprofloxacin against a resistant strain. In silico analysis also suggested favorable interactions between certain coumarins and the MepA efflux pump.

Summary: Cancer, a complex disease, is on the rise. Lignans, known for their anticancer properties, pose challenges in identifying specific subclasses with antitumor activity due to their structural diversity. This study aimed to explore the association between lignan subclasses and antitumor activity while considering the genetic profile of targeted variants. Predictive models were built for different targets, and molecular docking was performed to evaluate the binding energy of lignans with the best predicted biological activity. The results highlighted dibenzocyclooctadiene, furofuran, and aryltetralin subclasses as exhibiting strong anticancer activity.

Summary: The present study evaluated the antibacterial effect and inhibitory capacity of a, b, and d-damascone compounds against multidrug-resistant Staphylococcus aureus. These compounds showed clinically relevant effects against S. aureus standard strain and modulating effects with antibiotics against multidrug-resistant strains. They also demonstrated a modulating effect with antibiotics on efflux pumps, with d-damascone showing an inhibitory effect. In terms of ADME properties, a, b, and d-damascone exhibited favorable physicochemical properties, pharmacokinetic characteristics, and good interactions with the critical residues of the MepA receptor.

Summary: The study aimed to investigate the antibacterial and inhibitory activities of sesquiterpenes nerolidol, farnesol, and alpha-bisabolol, either individually or in liposomal nanoformulation, against multi-resistant Staphylococcus aureus strains carrying efflux systems NorA, Tet(K), MsrA, and MepA. The results showed that sesquiterpenes exhibited good antibacterial activity and efflux pump inhibition, while liposomal formulations had limited effectiveness.

The results showed that trans,trans-farnesol may have an antidepressant-like effect and does not have proconvulsant adverse effects.
