Local antibiotic delivery in periodontitis: drug release and its effect on supragingival biofilms

The effect of a drug-delivery system containing antibacterial metronidazole (MDZ) prescribed for periodontitis on supragingival biofilm was evaluated, and possible interference by this biofilm in the drug release profile was investigated. Streptococcus mutans biofilms were grown and exposed to a controlled-release formulation of MDZ or the same formulation without MDZ (vehicle control). Untreated biofilms were used as a negative control (NC). Biofilms and culture medium (containing detached cells) were collected 24, 48, 72, and 96 h after first exposure to treatments. The biomass of the MDZ group was lower than that of the NC group at all times. Although MDZ yielded low drug-release rates in the presence of the biofilm, it was sufficient for reducing viability for 24 h and affecting bacterial metabolism for 48 h. These results suggest that MDZ appears to destabilize supragingival biofilm. This biofilm may interfere with MDZ release from the formulation.

Summary: Candidiasis is a common opportunistic infection caused by fungi of the Candida genus, mainly affecting mucocutaneous tissues. Recurrent superficial candidiasis remains a challenge despite numerous available treatments, with biofilms playing a key role in antifungal resistance. Current research is focused on technological advancements in formulations to address the challenges, such as enhancing drug delivery system contact and permeation.

Synthesis and characterization of an experimental primer containing chitosan nanoparticles - Effect on the inactivation of metalloproteinases, antimicrobial activity and adhesive strength

Summary: The NPL-chitosan nanoparticles showed antibacterial activity against S. mutans, with MIC of 0.4% and MBC of 0.67%, and presented stable characteristics for at least 75 days. There was no statistical difference in Microtensile Strength among the experimental groups, but the group with 2% of NPL-chitosan exhibited higher MMP inactivation activity compared to others.

Summary: This study investigated the antimicrobial effect of anodic iontophoresis on Staphylococcus epidermidis and Escherichia coli. The results showed that anodic iontophoresis inhibited the growth of Staphylococcus epidermidis and caused death in Escherichia coli. It can be a powerful ally in controlling the proliferation of Escherichia coli in wounds.

Mutanase from Trichoderma harzianum inductively Produced by Mutan: Short-Term Treatment to Degrade Mature Streptococcus mutans Biofilm

Summary: This study evaluated the disruptive effect of fungal mutanase on cariogenic biofilm. Mature Streptococcus mutans biofilms were exposed to fungal enzymes for short-term treatment. The active enzymes significantly reduced insoluble polysaccharide content in the biofilm compared to the negative control group, without affecting bacterial viability and metabolism. These findings suggest that fungal mutanase could be used in oral care products.

Summary: Molar incisor hypomineralization (MIH) is often accompanied by dental hypersensitivity and difficulty in achieving effective analgesia. This study evaluated the effectiveness of preemptive analgesia in children with severe MIH, post-eruptive enamel breakdown, and hypersensitivity. The results showed that preemptive analgesia improved the effectiveness of infiltrative anesthesia and patient comfort during restorative procedures.

Needle-free anesthesia: clinical efficacy of a mucoadhesive patch for atraumatic anesthesia in dental procedures

Summary: This study demonstrates the clinical efficacy of mucoadhesive patches designed for needle-free pre-operative local anesthesia in dentistry. The patches were shown to initiate anesthesia within a short time frame, reaching maximum effect within 15 to 25 minutes, and lasting at least 50 minutes without any reported side effects.
