Summary: In this study, the effects of caspase-1 inhibitor VX765 on the expression of Cx43 and cell-to-cell communication after myocardial infarction (MI) were investigated. The results showed that VX765 treatment significantly decreased the infarct area, alleviated cardiac dysfunction and remodeling, and raised Cx43 levels in the heart after MI. In addition, VX765 treatment reversed the downregulation of Cx43 expression and improved intercellular communication in vitro.

Summary: The incidence and mortality of ventricular fibrillation (VF) among patients hospitalized for acute myocardial infarction (AMI) have significantly decreased over the past 20 years. However, VF remains associated with a 10-fold increased relative risk of in-hospital mortality, without affecting post-discharge mortality. These findings highlight the need for interventions during hospitalization to further reduce mortality in VF patients.

Results showed that voltage cut-offs applied to bipolar electrograms could delineate the scar, but not the VT circuit; electrogram fractionation had the highest correlation with scar transmurality; activation-repolarization interval identified regions closest to VT exit site but was outperformed by activation time gradients combined with voltage cut-offs; the performance of all metrics was affected by pacing location.

Summary: Connexin 43 (Cx43) plays an important role in conducting physiological responses in microvasculature. Previous studies have shown that the NADPH oxidase (NOX) enzymes, especially NOX2, are involved in cardiac redox biology and left ventricular remodeling after myocardial infarction. This study found that treatment with conjugated linoleic acid (cLA) significantly decreased survival in a murine model of myocardial infarction, possibly due to the disruption of Connexin 43 between myocytes in the ventricle. The mechanism underlying this effect involves the stimulation of NOX2 and the subsequent decrease in Cx43 levels, leading to gap junction dysfunction.

Summary: This study aimed to identify predictors of lethal ventricular arrhythmias (VAs) in patients with reduced left ventricular ejection fraction (LVEF) after acute myocardial infarction (AMI). The results showed that age and LVEF at admission were predictors of lethal VAs, with LVEF at admission being an independent predictor. This suggests that reduced LVEF and cardiac function at admission play a significant role in the risk stratification for future lethal VAs in this population.
