Cancer-associated fibroblasts secrete Wnt2 to promote cancer progression in colorectal cancer

Recent studies have shown that the tumor microenvironment plays a significant role in the progression of solid tumors. As an abundant component of the tumor microenvironment, cancer-associated fibroblasts (CAFs) have been shown to promote tumorigenesis and cancer aggressiveness, but their molecular characteristics remain poorly understood. In the present study, paired CAFs and normal fibroblasts (NFs) were isolated from five colorectal cancer (CRC) tissues from patients who underwent surgical resection. The gene expression profiles of CAFs and NFs identified by RNA sequencing were compared to understand the complex role of CAFs in cancer progression. Gene Set Enrichment Analysis revealed that the gene sets related to the Wnt signaling pathway were highly enriched in CAFs, as well as TGF beta signaling, which is considered to be a regulator of CAFs. Among the components of this pathway, Wnt2 was specifically expressed. The observations led us to speculate that Wnt2 is extremely involved in regulating CRC progression by CAFs. Thus, we performed immunohistochemical analysis on Wnt2 in 171 patients who underwent surgery for colorectal adenocarcinoma. Positive staining for Wnt2 was mainly observed in cancer stroma, although the immunoreactivity was weak in cancer cells. Wnt2 expression in CAFs was significantly correlated with depth of tumor (P < .001), lymph node metastasis (P = .044), TNM stage (P = .010), venous invasion (P < .001), and recurrence (P = .013). Subsequent in vitro analyses were conducted using conditioned medium (CM) from immortalized CAFs transfected with siRNA targeting Wnt2. As a result, cancer cell invasion and migration were significantly decreased in the CM from immortalized CAFs transfected with siRNA targeting Wnt2. Our findings indicated that Wnt2 protein released from CAFs enhances CRC cell invasion and migration. In conclusion, Wnt2 secreted by CAFs plays a key role in cancer progression and is a potential therapeutic target for CRC.

Summary: This study compared the postoperative outcomes of minimally invasive surgery (MIS) with open surgery (OS) in adult patients with congenital biliary dilatation (CBD). The results showed that MIS had significantly higher rates of type I CBD, significantly less blood loss, significantly lower rates of internal stents, significantly longer operation times, and significantly shorter postoperative hospital stays compared to OS. The postoperative outcomes of MIS for type I CBD were similar to those of the whole cohort. There were no significant differences in late complications between the groups. Robotic surgery had a significantly shorter operative time for hepaticojejunostomy (HJ) compared to laparoscopic surgery.

Summary: Recent advances in RNA studies have identified a novel estrogen-inducible antisense RNA, BNAT1, in ER-positive breast cancer. Silencing of BNAT1 inhibited the growth of tamoxifen-resistant breast cancer cells, repressed cell cycle progression, and promoted apoptosis. BNAT1 may play a critical role in ER-positive breast cancer biology by modulating ER alpha-dependent transcription regulation.

The Pro-Tumorigenic Role of Chemotherapy-Induced Extracellular HSP70 from Breast Cancer Cells via Intratumoral Macrophages

Summary: Tumor-associated macrophages (TAMs) contribute to tumor progression and chemoresistance, so it is important to clarify the altered functions of macrophages following chemotherapy. This study investigated the effects of heat shock protein 70 (HSP70) secretion from breast cancer cells on macrophage function. The results showed that extracellular HSP70 levels increased following chemotherapy, enhancing the pro-tumorigenic effects of TAMs by regulating transforming growth factor (TGF)-beta expression in breast cancer cells. HSP70 was found to be a poor prognostic factor associated with macrophage infiltration.

New Perspectives on Sex Steroid Hormones Signaling in Cancer-Associated Fibroblasts of Non-Small Cell Lung Cancer

Summary: The importance of sex hormones, especially estrogen, in the pathogenesis of non-small cell lung cancer (NSCLC) has attracted attention due to its high incidence among young adults and nonsmokers, especially women. Cancer-associated fibroblasts (CAFs), which reside in the cancer stroma, play a significant role in cancer growth, invasion, metastasis, and drug resistance through interactions with cancer cells and other microenvironmental components. However, the importance of sex hormones in the cell-cell interactions of CAFs in NSCLC remains unclear.

Infliximab Inhibits Colitis Associated Cancer in Model Mice by Downregulating Genes Associated with Mast Cells and Decreasing Their Accumulation

Summary: This study aimed to investigate the potential of anti TNF-alpha antibodies (Abs) in inducing colon cancer development and identify the genes involved in colitis-associated cancer. The results showed that TNF-a inhibited the proliferation, migration, and invasion of colon cancer cell lines, and the neutralization of TNF-a by anti TNF-a Abs was observed in vitro. In a mouse model of colitis-associated cancer, treatment with infliximab, an anti TNF-a Ab, significantly reduced the development of colon cancer. Furthermore, gene analysis revealed the role of mast cells in the development of cancer associated with colitis.
