HER2 Regulates Cancer Stem Cell Activities via the Wnt Signaling Pathway in Gastric Cancer Cells

Introduction: Human epidermal growth factor 2 (HER2) gene overexpression in breast carcinoma cell lines has been shown to drive mammary carcinogenesis and tumor growth and invasion through its effects on mammary stem cells. Objective: Therefore, we investigated the mechanism by which HER2 regulates cancer stem cell (CSC) activity in gastric cancer cells. Methods:HER2 was transfected into MKN28 gastric cancer cells, and its role in regulating CSC activity was determined by characterizing the HER2-overexpressing cells. Results: The sphere formation assay revealed that the sphere sizes and frequency of sphere formation were significantly greater for the HER2-overexpressing cells than for the MKN28 control cells. The CSC markers Oct-4 and BMI1 were more highly expressed in the HER2-overexpressing cells, as were the EMT markers. This was accompanied by a significant enhancement in cellular invasion of the Matrigel and migration. The E-cadherin level was significantly downregulated, and the mesenchymal marker Snail upregulated, in the HER2-transfected cells. HER2 overexpression activated the well-characterized CSC-associated Wnt/beta -catenin signaling pathway, as shown by the luciferase assay. After treatment of these cells with the Wnt signal inhibitor PRI-724, the BMI1 and Oct-4 levels were decreased for 24 h and Snail was also downregulated. Immunofluorescence staining revealed the significant restoration of E-cadherin levels in the HER2-transfected cells after PRI-724 treatment. Conclusions: These results established a role for HER2 in regulating gastric CSC activity, with Wnt/beta -catenin signaling being mediated via a HER2-dependent pathway. In summary, HER2-overexpressing gastric cancer cells exhibited increased stemness and invasiveness and were regulated by Wnt/beta -catenin signaling.

Summary: This study investigated the impact of different doses of carboplatin on pathologic complete response rate, grade 3/4 anaemia occurrence, and transfusion rate in patients treated with neoadjuvant TCHP. The results showed that carboplatin at AUC5 has comparable cytotoxic effects to carboplatin at AUC6 and is associated with fewer complications related to anaemia in HER2+ breast cancer patients.

Pseudomonas stutzeri PM101005 inhaled with atmospheric particulate matter induces lung damage through inflammatory responses

Summary: In this study, it was found that PM-isolated Pseudomonas stutzeri (PMPS) induces higher levels of inflammatory responses in lung epithelial cells, alveolar cells, and macrophages compared to Pseudomonas stutzeri (PS). The pathogenicity of PMPS is mediated by Toll-like receptors (TLRs) signaling pathways, which also regulate the expression of the antibiotic peptide beta-defensin 3 (BD3) that suppresses PMPS proliferation. Furthermore, pretreatment with PM enhances inflammatory responses and tissue damage caused by PMPS, while reducing BD3 expression.

Relieving Anxiety Through Virtual Reality Prior to Endoscopic Procedures

Summary: This study found that exposing patients to virtual reality (VR) prior to endoscopic procedures can reduce their anxiety levels, leading to less discomfort and incomplete procedures, as well as decreased use of sedative medication. The proportion of patients with high anxiety significantly decreased in the VR group compared to the control group, and patients in the VR group reported higher satisfaction with sedation.

Potential Association of Osteoporosis and Not Osteoporotic Fractures in Patients with Gout: A Longitudinal Follow-Up Study

Summary: Using nationwide data from the Korean National Health Insurance Service-Health Screening Cohort database, this study aimed to explore the link between gout and incident osteoporosis/osteoporotic fractures. The results showed that gout patients had a slightly higher risk of developing osteoporosis compared to controls, but no significant association was found between gout and incident osteoporotic fractures. This study provides insights into the health implications of gout in the Korean population.

A Comparative Analysis of Bi-Portal Endoscopic Spine Surgery and Unilateral Laminotomy for Bilateral Decompression in Multilevel Lumbar Stenosis Patients

Summary: In this retrospective study, the clinical and radiological outcomes of patients with multilevel lumbar stenosis who underwent BESS and microscopic ULBD were compared. The results showed that the BESS group had better clinical outcomes, with significant improvements in back and leg pain scores and medication use. Radiological outcomes were also superior in the BESS group. The short-term follow-up period demonstrated the efficacy of multilevel decompression with the BESS technique.

Electrical role of sulfur vacancies in MoS2: Transient current approach

Summary: The study investigates the effect of sulfur vacancies on the electrical properties of MoS2. After generating sulfur vacancies through treatment with hydrazine, the density of shallow traps increased by a factor of 4, whereas that of deep-level traps increased by a factor of 200. These results indicate that sulfur vacancies induce both deep- and shallow-level traps, with a higher trap density at the deep level.

Nomogram for pre-procedural prediction of non-curative endoscopic resection in patients with early gastric cancer

Summary: By analyzing the characteristics of patients with early gastric cancer who underwent ESD, we developed a nomogram to predict the risk of non-CR. This nomogram includes endoscopic features, radiologic findings, and pathology results, and it showed good predictive performance in the external validation.

Angiotensin II-Related Activation of Scleral Fibroblasts and Their Role on Retinal Ganglion Cell Death in Glaucoma

Summary: We investigate the changes in extracellular matrix (ECM) and biomechanical properties of the sclera after systemic hypotension induced by oral hydrochlorothiazide. Angiotensin II (AngII) receptor levels and ECM components were evaluated, along with the effect of losartan inhibition and its impact on retinal ganglion cells (RGCs). The results show that AngII plays a role in scleral fibrosis after systemic hypotension, and inhibiting AngII could modulate the tissue properties of the sclera, providing protection for RGCs.

Risk of Metabolic Syndrome and Fatty Liver Diseases in Gastric Cancer Survivors: A Propensity Score-Matched Analysis

Summary: This study aimed to compare the risk of metabolic syndrome and fatty liver diseases between gastric cancer survivors and non-cancer subjects. The results showed that gastric cancer survivors who underwent surgical treatment had lower risks of metabolic syndrome, fatty liver by ultrasonography, and metabolic dysfunction-associated fatty liver disease (MAFLD) than non-cancer subjects. However, there were no significant differences in the risks between non-surgical gastric cancer survivors and non-cancer subjects.

Summary: This study analyzed data from postmenopausal women with HR-positive, HER2-negative ABC who were treated with CDK4/6 inhibitors and found that on-treatment dNLR predicted progression-free survival (PFS). Flow cytometry analysis revealed potential systemic immune responses induced by CDK4/6 inhibitors, including prevention of T cell exhaustion and reduction of myeloid-derived suppressor cell frequency.
