Summary: The study showed that upregulation of AHNAK2 in thyroid carcinoma tissues, especially in metastatic ones, is associated with poor prognosis. Knockdown of AHNAK2 inhibited thyroid carcinoma migration, invasion, and metastasis by reducing activation of the NF-kappa B pathway. This provides a novel target for treating metastatic thyroid carcinoma.

Summary: This study confirmed that cytoplasmic protein TRAF2 can be methylated by SMYD2 and contribute to the continuous activation of NF-kappa B signaling pathway. Knocking down SMYD2 showed varying degrees of mitigation in acute and chronic inflammation mouse models. Moreover, lysine-specific demethylase LSD1 was found to resist the methylation of TRAF2 induced by SMYD2.

UCHL3 stabilizes YAP through its deubiquitylation activity, and its depletion leads to decreased ATC progression, stemness, metastasis, and increased sensitivity to chemotherapy. The study also revealed that TEAD4, which binds to YAP and DNA, activates UCHL3 transcription by binding to its promoter.

Summary: TRAF4 is overexpressed in glioblastoma and is associated with glioma subtype and grade. Knockdown of TRAF4 inhibits the growth and invasion of glioblastoma cells. TRAF4 mediates the ubiquitination of SETDB1 to maintain its stability and function, thereby promoting the activation of the AKT pathway. Restoring SETDB1 expression partially rescues the growth and proliferation of TRAF4 knockdown glioblastoma cells.
