Summary: This study compares the performance of three commercial kits in detecting CEBPA and FLT3 mutations. The results show that all three kits have 100% sensitivity in detecting CEBPA and FLT3 mutations, and there is high concordance in detecting confirmed variants in core myeloid genes. These findings indicate that NGS assays can now reliably detect CEBPA and FLT3 mutations.

Summary: This study introduces a novel and efficient FLT3-ITD detection approach called ITDetect, which utilizes NGS data. ITDetect demonstrates higher accuracy and provides more detailed information compared to existing in silico methods. Validation experiments show that ITDetect has the highest concordance with experimental methods.

Summary: The aim of the study was to compare the effect of Quizartinib versus placebo on overall survival in patients with FLT3-ITD-positive newly diagnosed AML. The results showed that the addition of Quizartinib to standard chemotherapy can improve overall survival in adult patients with FLT3-ITD-positive newly diagnosed AML.

Summary: The study investigated the feasibility and efficacy of adding sorafenib, a multikinase tyrosine kinase inhibitor, to standard chemotherapy and as single-agent maintenance therapy in pediatric acute myeloid leukemia (AML) with high allelic ratio (HAR) FLT3/ITD mutations. The results showed that sorafenib can be safely added to conventional AML chemotherapy and may improve outcomes in this population. Patients treated with sorafenib had higher survival rates and disease-free survival rates compared to those who did not receive sorafenib.

Summary: The study found that FLT3-ITDlow in AML patients is associated with negative enrichment of leukemic stem cell features, marked enrichment of the RAS pathway, and higher frequencies of RAS pathway mutations, different from FLT3-ITDhigh. Concurrent CEBPA double mutations are favorable prognostic factors, while MLL-PTD and mutations in splicing factors are unfavorable prognostic factors in FLT3-ITDlow patients. Allo-HSCT in CR1 significantly prolongs overall survival and event-free survival in FLT3-ITDlow patients.

Summary: Patients with large granular lymphocytic leukemia (LGLL) frequently present with neutropenia. This study focuses on LGLL patients with isolated anemia, which is uncommon. It found that male patients were more likely to have isolated anemia, and those with pure red cell aplasia (PRCA) required more transfusion-dependence. Treatment resulted in a good overall response rate, and there was no significant difference in overall survival between PRCA and non-PRCA patients.

Summary: Therapy-related myeloid neoplasms (t-MN) are aggressive forms of leukemia that occur as a complication of prior exposure to DNA-damaging agents. Clonal cytopenia of undetermined significance (CCUS) is a precursor to these neoplasms. This study identified distinct characteristics and outcomes associated with therapy-related clonal cytopenia (t-CC), providing insights into the development and progression of t-MN.

Summary: Von Willebrand factor (VWF) is an important indicator for heritable bleeding disorders. Newer assays for assessing VWF platelet-binding activity have been recommended, but their availability is limited in the United States. This study aims to evaluate the utilization trends, agreement of testing methods, and imprecision of VWF testing.

Summary: Inherited platelet disorders (IPDs) are a diverse group of disorders with normal or reduced platelet counts, varying severities of bleeding tendencies, and involvement or non-involvement of other organs. Diagnosing the underlying cause of IPDs is challenging due to the lack of highly specific platelet function tests and overlapping clinical and laboratory features. Genetic testing through next-generation sequencing (NGS) technologies has become an important part of the laboratory investigation for patients with IPDs, along with thorough clinical and laboratory testing and phenotype and genotype correlation studies.

Summary: The 2022 ICC and WHO 5th edition classifications have proposed significant changes to the diagnostic criteria of MDS. This study evaluated their impact on the diagnosis of cytopenias and MDS. The results showed major changes in the diagnosis according to both classification systems, with some differences in reclassification of specific MDS entities. It is important to assess the potential diagnostic and clinical impacts before implementing these new classification systems.

Summary: Computational pathology research driven by deep learning faces challenges in reproducibility and reusability. Codebase with good documentation and robustness and generalizability of models are crucial. The reuse of computational pathology algorithms is limited, and their application in clinical settings is even rarer. This study evaluates 160 peer-reviewed articles, providing criteria for data and code availability and statistical analysis of results.

Summary: The Amsterdam Consensus Statement introduced the term maternal vascular malperfusion (MVM) to classify a group of findings related to impaired maternal-placental circulation. The study found that features such as low placental weight, accelerated villous maturation, decidual arteriopathy, and infarcts are associated with adverse obstetrical outcomes, while the role of other features like distal villous hypoplasia, excess multinucleated trophoblast, and retroplacental hemorrhage needs further research.
