Pregnancy-Associated Plasma Protein A Induces Inflammatory Cytokine Expression by Activating IGF-I/PI3K/Akt Pathways

Pregnancy-associated plasma protein A (PAPP-A) was previously reported to be an inflammatory biomarker and a prognostic marker of acute coronary syndrome (ACS) and involved in the process of atherosclerosis and plaque rupture. However, the role of PAPP-A in inflammation is poorly understood. In this study, we aimed to investigate the role of PAPP-A in macrophage activation and inflammatory cytokine production. RAW264.7 macrophages were treated with or without PAPP-A. Reverse-transcriptase quantitative real-time PCR (RT-qPCR) and Western blot were performed to detect gene and protein expressions. The concentration of monocyte chemoattractant protein-1 (MCP-1), tumor necrosis factor-alpha (TNF-alpha), and interleukin-6 (IL-6) in culture supernatants was determined by ELISA. Results showed that PAPP-A significantly stimulated the expression of MCP-1, TNF-alpha, and IL-6 at both transcriptional and translational levels in a dose-dependent and time-dependent manner. The secretion of these inflammatory cytokines by macrophages was also increased after PAPP-A treatment. Moreover, PAPP-A activated the IGF-I/PI3K/Akt signaling pathway in macrophages. The PAPP-A-mediated upregulation of MCP-1, TNF-alpha, and IL-6 mRNA and protein levels were strongly inhibited by PI3K inhibitors or IGF-IR siRNA, indicating that the upregulation of MCP-1, TNF-alpha, and IL-6 could involve the IGF-I/PI3K/Akt pathway. Together, this study demonstrates that PAPP-A activates the macrophage signaling pathway (IGF-I/PI3K/Akt), which drives the expression and production of inflammatory cytokines known to contribute to the initiation and progression of ACS. These findings indicate that PAPP-A may play a proinflammatory role in the pathophysiology of ACS and serve as a potential therapeutic target.

OLIF versus ALIF: Which is the better surgical approach for degenerative lumbar disease? A systematic review

Summary: The study compared the clinical and radiological outcomes of OLIF and ALIF for treating lumbar degenerative diseases. The results showed no significant difference in disc height, segmental lordosis angle, lumbar lordosis, VAS score, and ODI between the two groups. Operation time, blood loss, and hospital stay were comparable. Both groups achieved over 90% fusion rate, but OLIF had a higher complication rate, particularly cage subsidence. In conclusion, OLIF and ALIF are both effective treatments with similar therapeutic effects, but ALIF may be more costly for patients due to the involvement of vascular surgeons.

Accuracy of cortical bone trajectory screw fixation guided by spinous process clamp guide in lumbosacral vertebrae: A cadaver study

Summary: This study evaluated the accuracy of cortical bone trajectory screw placement guided by spinous process clamp. Using 8 lumbar specimens, a total of 96 screws were implanted. The results showed that the SPC guide group had higher clinically acceptable placement and reduced severe deviations in important directions. Therefore, implanting CBT screws guided by SPC can improve surgical accuracy.

A numerical study on damage characteristics in composite tapered laminates under cyclic loading with different stress ratios

Summary: The design of tapered composite fan blades with increased thickness near the root helps carry the load, but also introduces material and geometric discontinuities that can lead to interlayer stresses and damage initiation. A study using high-fidelity finite element modeling and a constitutive equation found that the maximum damage index occurs at the ply interface and resin pocket of the laminates. Static damage and weak-link index assessment were proposed to predict the location of fatigue damage. The study also found that fatigue crack growth rates are sensitive to cyclic loading at the first natural frequency and the weak-link zone appears in the crack growth area.

Experimental Study on Grouting Diffusion Law of the Different Crack Widths in Tunnel Lining

Summary: This study investigated the diffusion shape and the relation between diffusion radius and time of grouting in tunnel lining cracks. The diffusion equation of grouting in lining cracks was established, and the correlation between experimental and calculated values was analyzed. The results showed that the diffusion shape in tunnel lining cracks can be categorized into circular, excessive, and elliptical zones, and the diffusion equation accurately describes the relationship between diffusion radius and time. These findings provide a theoretical basis for predicting diffusion radius in grouting repair for tunnel lining cracks.

Prevalence and antimicrobial resistance of Salmonella and the enumeration of ESBL E. coli in dairy farms in Hubei Province, China

Summary: The prevalence and antimicrobial resistance of Salmonella and ESBL-producing E. coli in healthy lactating dairy cows in Hubei Province, China was investigated in this study. Salmonella was found in 5.2% of sampled cows, with a herd prevalence of 20.0%. ESBL E. coli was detected in all farms, with an animal level prevalence of 59.1% and more than 104 CFU/g of feces. These findings provide valuable information for controlling Salmonella in Hubei Province.

Is It Necessary to Cross the Cervicothoracic Junction in Posterior Cervical Decompression and Fusion for Multilevel Degenerative Cervical Spine Disease? A Systematic Review and Meta-Analysis

Summary: This study compared the outcomes of posterior cervical decompression and fusion (PCF) terminating at the lower cervical spine and crossing the cervicothoracic junction (CTJ). The results showed that terminating at the lower cervical spine was associated with fewer wound-related complications, lower reoperation rate for wound-related complications, and less neck pain at the final follow-up. However, it also had a higher incidence of overall adjacent segment disease and hardware failure. The operating time and estimated blood loss were shorter when the PCF construct did not cross the CTJ.

Biomechanical evaluation of multiple pelvic screws and multirod construct for the augmentation of lumbosacral junction in long spinal fusion surgery

Summary: This study compared the biomechanical performance of combining multiple pelvic screws and multirod construct to other advanced sacropelvic fixation constructs for the augmentation of lumbosacral junction in long spinal fusion surgery through finite element analysis. The results showed that both multiple pelvic screws and multirod construct could increase the mechanical stability of lumbosacral junction and reduce stress on instrumentation, cages, S1 superior endplate, and sacrum. The MS-MR construct was the most adequate to reduce the risk of lumbosacral pseudarthrosis, implant failure, and sacrum fracture. This study provides important evidence for the application of MS-MR construct in clinical settings.

A new type of generic, self-evolving and efficient automated deduction algorithm based on category theory

Summary: This article presents a new type of automated statement proof algorithm that is generalized, self-evolving, and efficient, using new data structures (brackets and map graphs) and algorithms. The brackets structure provides a concise representation of mathematical concepts, while the map graphs enable the computer to learn while proving using an efficient machine-learning method. Additionally, the algorithm is based entirely on category theory. Finally, a prototype of the program is introduced and its performance examined.

Best Practice Guide for Cryoballoon Ablation in Atrial Fibrillation: The Compilation Experience of More than 1000 Procedures

Summary: Nowadays, cryoballoon (CB) has become an established alternative to radio frequency (RF) ablation for pulmonary vein isolation (PVI), with the ability to isolate PVs in a single application. In a study on over 1000 consecutive patients, our center optimized the CB PVI procedure. It is expected that future guidelines will recommend CB as the first-line PVI for patients with paroxysmal AF and a class I indication. In a long-term follow-up of the EARLY-AF trial, CB showed a lower incidence of persistent atrial fibrillation compared to anti-arrhythmic drugs.

Transcriptomic analysis of Listeria monocytogenes biofilm formation at different times

Summary: This study used transcriptomics to analyze differential gene expression between biofilm and planktonic bacteria at different stages of biofilm formation and growth in Listeria monocytogenes. They identified a significant number of differentially expressed genes and demonstrated changes associated with various biological processes and pathways. The expression of the top upregulated and downregulated genes was validated through real-time quantitative polymerase chain reaction. These findings lay the foundation for further research on biofilm formation mechanisms and the development of biofilm inhibitors to reduce LM infection risk.
