Three Discrete Patterns of Primary Aldosteronism Lateralization in Response to Cosyntropin During Adrenal Vein Sampling

Context: Cosyntropin [ACTH (1-24)] stimulation during adrenal vein (AV) sampling (AVS) enhances the confidence in the success of AV cannulation and circumvents intraprocedure hormonal fluctuations. Cosyntropin's effect on primary aldosteronism (PA) lateralization, however, is controversial. Objectives: To define the major patterns of time-dependent lateralization, and their determinants, after cosyntropin stimulation during AVS. Methods: We retrospectively studied patients with PA who underwent AVS before, 10, and 20 minutes after cosyntropin stimulation between 2009 and 2018. Unilateral (U) or bilateral (B) PA was determined on the basis of a lateralization index (LI) value >= 4 or < 4, respectively. Available adrenal tissue underwent aldosterone synthase-guided next-generation sequencing. Results: PA lateralization was concordant between basal and cosyntropin-stimulated AVS in 169 of 222 patients (76%; U/U, n = 110; B/B, n = 59) and discordant in 53 patients (24%; U/B, n = 32; B/U, n = 21). Peripheral and dominant AV aldosterone concentrations and LI were highest in U/U patients and progressively lower across intermediate and B/B groups. LI response to cosyntropin increased in 27% of patients, decreased in 33%, and remained stable in 40%. Baseline aldosterone concentrations predicted the LI pattern across time (P < 0.001). Mutation status was defined in 61 patients. Most patients with KCNJ5 mutations had descending LI, whereas those with ATP1A1 and ATP2B3 mutations had ascending LI after cosyntropin stimulation. Conclusion: Patients with severe PA lateralized robustly regardless of cosyntropin use. Cosyntropin stimulation reveals intermediate PA subtypes; its impact on LI varies with baseline aldosterone concentrations and aldosterone-driver mutations.

Personalized Treatment of Patients With Primary Aldosteronism

Summary: Primary aldosteronism (PA) is a common but underestimated secondary cause of hypertension, associated with increased cardiovascular and renal morbidity compared to primary hypertension. Early identification and targeted therapy are crucial in reducing the morbidity and mortality in a large hypertensive population. Unilateral adrenalectomy is the preferred treatment for lateralized PA, while medical therapy with mineralocorticoid receptor antagonists is offered to bilateral PA patients and those who are not surgical candidates. Novel therapies are being developed as alternative options for PA treatment. This review article discusses the individualized therapy for patients with PA.

Boronate Avidity Assisted by Dendrimer-like Polyhedral Oligomeric Silsesquioxanes for a Microfluidic Platform for Selective Enrichment of Ubiquitination and Glycosylation

Summary: A new strategy utilizing a dendrimer-like structure of boron materials based on octavinyl-polyhedral oligomeric silsesquioxanes (Ov-POSS) was suggested to improve boronate avidity with good accessibility of sites. The resulting Ov-POSS cross-linked boron monolith showed significantly stronger affinity for glycoproteins compared to the Ov-POSS-free monolith. The developed microfluidic platform, which integrated multiple functions, demonstrated high efficiency and high-throughput analysis for post-translational modification proteomics.

Daurisoline suppresses esophageal squamous cell carcinoma growth in vitro and in vivo by targeting MEK1/2 kinase

This study investigated the effects of Daurisoline, a natural compound, on ESCC and identified its mechanism of action. Daurisoline inhibited ESCC growth by suppressing the ERK1/2 signaling pathway, making it a potential MEK inhibitor for ESCC treatment.

Effects of simulated drought stress on the growth and physiological and biochemical parameters of Paspalum wettsteinii

Summary: Paspalum wettsteinii shows adaptive responsiveness to drought stress, with mild drought stress improving growth and photosynthesis, while extreme drought stress decreasing physiological and biochemical parameters. Moderate and severe drought stress responses depend on exposure time. Short-term drought stress enhances photosynthesis and antioxidant capacity, but these parameters are inhibited with longer exposure time.

A Glycosidic-Bond-Based Mass-Spectrometry-Cleavable Cross-linker Enables In Vivo Cross-linking for Protein Complex Analysis

Summary: Chemical cross-linking mass spectrometry (CXMS) is a powerful technology for analyzing protein complexes. However, in vivo CXMS studies have been limited by cross-linking biocompatibility and data analysis. In this study, a glycosidic bond-based MS-cleavable cross-linker called trehalose disuccinimidyl ester (TDS) was designed and synthesized, which simplified the cross-linked peptides into conventional single peptides via selective cleavage. TDS showed high biocompatibility and accuracy, enhancing the cross-linking identification accuracy and throughput.

Enhanced energy storage performance of Sr0.7Bi0.2TiO3@ NaNb0.9Ta0.1O3 relaxor ferroelectrics via a synergistic optimization strategy

Summary: In this study, Sr0.7Bi0.2TiO3-based ceramics with excellent dielectric temperature stability and energy storage efficiency were prepared by a two-step sintering process. The introduction of NNT and the design of a core-shell structure enhanced the relaxation behavior and temperature stability of the ceramics. The obtained ceramics showed promising energy storage performance and could be applied in dielectric ceramic capacitors.

Apple polyphenol biotransformation using probiotics in vitro and dynamic simulated digestion by bionic rats

Summary: This study investigated the effects of fermentation and dynamic simulated digestion on the biotransformation and antioxidant potential of apple polyphenols. The polyphenol content of fermented apple pulp was significantly higher compared to non-fermented pulp, and the antibacterial activity improved after fermentation. The antioxidant potential of apple pulp also increased after fermentation and digestion. The findings suggest that fermented and digested apple polyphenols have better bioavailability and can be beneficial for the absorption of active substances.

Interplay of Cortisol, Testosterone, and Abdominal Fat Mass in Normal-weight Women With Polycystic Ovary Syndrome

Summary: Ovarian and adrenal steroidogenesis play a role in endocrine-metabolic dysfunction in polycystic ovary syndrome (PCOS). Comparing PCOS patients with normal-weight controls, elevated serum levels of total/free testosterone (T) and androstenedione (A4) were observed in PCOS patients, along with greater android/gynoid fat mass. Adrenal steroids were not significantly associated with body fat distribution, but reduced cortisol levels correlated with diminished android fat mass in PCOS patients, suggesting a protective effect against preferential abdominal fat deposition.

Glucose metabolism impairment in Parkinson's disease

This review discusses the glucose metabolism impairment in PD, its pathophysiological mechanisms, and available therapies targeting glucose metabolism impairment in PD.

Brazilin Inhibits the Invasion and Metastasis of Breast Cancer

Summary: This study aimed to investigate the effect of brazilin on breast cancer invasion and metastasis. The results showed that brazilin inhibited the proliferation and invasion of breast cancer cells, as well as significantly decreased the number of metastatic nodules in the lungs. In addition, brazilin also increased the average survival days and spleen index in the mice. These findings suggest that brazilin may have potential as a new therapeutic agent for breast cancer.

MVFI-Net: Motion-Aware Video Frame Interpolation Network

Summary: Video frame interpolation (VFI) aims to generate intermediate frames between successive frames. Existing learning-based methods often suffer from limited reference regions and complex motions. This paper proposes a novel motion-aware VFI network (MVFI-Net) which overcomes these issues by introducing a motion-aware convolution operation and incorporating pyramid structure. Experimental results demonstrate that the proposed method achieves state-of-the-art performance on diverse benchmarks.
