Synergistic anti-breast cancer effect of pulsatilla saponin D and camptothecin through interrupting autophagic-lysosomal function and promoting p62-mediated ubiquitinated protein aggregation

Autophagy is an evolutionarily conserved mechanism to protect the cells from unfavorable environmental conditions. Inhibition of autophagy has been contemplated as a novel strategy to enhance anticancer efficacy of existing chemotherapeutic agents. We previously reported that pulsatilla saponin D (PSD) was a potent autophagy inhibitor. However, its anticancer potential as adjuvant and underlying mechanisms are still unknown. In this study, we identified that PSD induced the formation of autophagosome in MCF-7 and MDA-MB-231 breast cancer cells. However, PSD alone and particularly co-treatment with camptothecin remarkably increased p62 protein levels, indicating that PSD strongly inhibited the autophagic cargo degradation. The mechanistic study indicated that PSD profoundly abolished the co-localization of EGFP-LC3 and lysosomal-specific probe LysoTracker Red, suggesting that the autophagosomelysosome fusion was blocked by PSD, which is similar to the action of chloroquine. In addition, PSD significantly increased lysosomal pH and inhibited the activation of lysosomal cathepsins in both breast cancer cell lines. Furthermore, the accrued p62 resulted in accumulation of ubiquitinated proteins owing to the interaction with p62 and delivery to the malfunctioned autophagosome by PSD. Finally, we demonstrated that PSD synergistically enhanced the anticancer activity of camptothecin (CPT) in cultured breast cancer cells and in mouse xenograft tumor models. Our results indicated that PSD inhibited autophagic flux via blocking autophagosome-lysosome fusion and lysosomal acidification, which may confer a synergistic anti-breast cancer activity of PSD and CPT.

Characterizing the influence of different drying methods on chemical components of Panax notoginseng leaves by heart-cutting two-dimensional liquid chromatography coupled to orbitrap high-resolution mass spectrometry

Summary: The study found that different drying methods significantly influence the chemical components in Panax notoginseng leaves (PNL), with sun-air drying and hot-air drying leading to vigorous ginsenoside transformation. Specifically, the main components of PNL can be transformed into different compounds by selective cleavage of beta-1,2-glucosidic linkage, with only steaming prior to drying able to inhibit this transformation. These findings provide a scientific basis for designing industrial drying methods to ensure PNL quality.

Red yeast rice ameliorates non-alcoholic fatty liver disease through inhibiting lipid synthesis and NF-κB/NLRP3 inflammasome-mediated hepatic inflammation in mice

RYR reduces lipid accumulation and hepatic inflammation, regulates lipid metabolism, and inhibits the inflammatory response in mice and cell models.

Integrative computational approach identifies immune-relevant biomarkers in ulcerative colitis

Summary: Ulcerative colitis is a complex inflammatory bowel disease with a genetic and immune etiology. This study identified potential biomarkers for ulcerative colitis and analyzed immune cell infiltration characteristics using bioinformatic analyses. The differentially expressed mRNAs (DEMs) were mainly enriched in infiltrating neutrophils and regulation of leukocyte migration. Specifically, three candidate biomarkers were strongly correlated with infiltrating immune cells of ulcerative colitis. These findings suggest the importance of these genes as diagnostic biomarkers and the involvement of immune infiltration cells in the progression of ulcerative colitis.

Integrating diverse plant bioactive ingredients with cyclodextrins to fabricate functional films for food application: a critical review

Summary: The popularity of plant bioactive ingredients in the food industry is increasing. However, these ingredients have some deficiencies. Cyclodextrins (CDs) have been extensively studied as excellent carriers for plant bioactive ingredients and can be added to film materials to create bioactive food packaging. This review summarizes the extraction methods of plant bioactive ingredients, the addition of CD inclusion compounds to thin-film materials, and their applications in food packaging. The release model and mechanism of active film materials based on various plant bioactive ingredients with CDs are also highlighted.

Chemical Stability of a Chinese Herbal Spirit Using LC-MS-Based Metabolomics and Accelerated Tests

Role of the Epigenetic Modifier JMJD6 in Tumor Development and Regulation of Immune Response

Abnormal expression of JMJD6 is associated with various tumors and it also regulates the immune response. Targeting the JMJD6 gene may be a promising strategy for cancer therapy.

Non-small-cell lung cancer patients harboring TP53/KRAS co-mutation could benefit from a PD-L1 inhibitor

Summary: This study explored the association between TP53 mutation and atezolizumab in non-small-cell lung cancer (NSCLC) patients. It was found that NSCLC patients with TP53/KRAS co-mutation had longer overall survival (OS) with atezolizumab compared to docetaxel. However, there was no significant difference in OS between atezolizumab and docetaxel for NSCLC patients with only TP53 mutation or KRAS mutation.

Biopolymer Food Packaging Films Incorporated with Essential Oils

Summary: Petroleum-based packaging materials are harmful to the environment and human health due to their nonbiodegradable nature. Researchers are now focusing on developing efficient, biodegradable, and nontoxic food packaging films. Essential oils (EOs) are being used as a natural alternative to synthetic additives due to their favorable antioxidant and antibacterial properties. This review summarizes the mechanisms of EOs and analyzes the properties and performance of food packaging films incorporating EOs. The future prospects and challenges of using EOs-containing biopolymer films in food packaging are also discussed.

Antiviral Potential of Small Molecules Cordycepin, Thymoquinone, and N6, N6-Dimethyladenosine Targeting SARS-CoV-2 Entry Protein ADAM17

Summary: This study evaluated the inhibitory effects of cordycepin, thymoquinone, and N6, N6-dimethyladenosine on ADAM17 in cancer cells and predicted their potential in anti-COVID-19 therapy. The results showed that these compounds suppressed the expression of ADAM17 and had potential against SARS-CoV-2. Pharmacological analysis indicated that ADAM17 might be a target for anti-COVID-19 therapy with these compounds.

Immunomodulatory role of metalloproteases in cancers: Current progress and future trends

Summary: Metalloproteinases (MPs) are a family of proteinases that contain metal ions in their active centers. They play various roles in cancer, including protein hydrolysis, cell adhesion, and remodeling of the extracellular matrix. MPs are expressed in multiple types of cancers and participate in pathological processes involving tumor development, invasion, and metastasis by regulating signal transduction and the tumor microenvironment. In this review, we summarize the structure of MPs, their expression, and especially their immunomodulatory role and mechanisms in cancers. We also provide an update on recent advances and future directions in the diagnosis and immunotherapy targeting MPs in cancers.

Immunomodulatory role of metalloproteinase ADAM17 in tumor development

Summary: ADAM17 is a transmembrane protease involved in cell membrane protein shedding and signal pathway regulation. It has been shown to regulate over 90 substrates, some of which play important roles in tumor formation and development. ADAM17 also has immunomodulatory roles, although the mechanisms are still not well understood. This paper provides a summary of the structure, regulatory roles, and immunomodulatory functions of ADAM17, as well as the potential use of small molecule inhibitors and monoclonal antibodies for targeting ADAM17.

Whole exome sequencing in Chinese mucinous pulmonary adenocarcinoma uncovers specific genetic variations different from lung adenocarcinoma

Summary: This study describes the genomic and immunological landscape of Chinese mucinous pulmonary adenocarcinoma (MPA) and compares it with classical lung adenocarcinoma (LUAD). It is found that MPA has distinct genomic alterations and a more complex genomic profile compared to LUAD. Mutations in specific pathways are associated with survival outcomes, and mutations in immune-related genes also affect prognosis.

Ginsenoside Rk2, a dehydroprotopanaxadiol saponin, alleviates alcoholic liver disease via regulating NLRP3 and NLRP6 inflammasome signaling pathways in mice

Summary: The potential beneficial effects of ginsenoside Rk2 in mitigating alcoholic liver injury in mice are reported. Rk2 acts as an inhibitor of the NLRP3 inflammasome signaling in the liver, reducing liver inflammation caused by chronic-plus-single-binge ethanol feeding. Additionally, Rk2 improves alcohol-induced intestinal barrier dysfunction by enhancing NLRP6 inflammasome in the intestine.
