Summary: HIGD2A, an essential protein for mitochondrial respiratory supercomplex assembly, is overexpressed in hepatocellular carcinoma (HCC) and associated with poor prognosis. Knockdown of HIGD2A inhibits HCC cell proliferation and migration, arrests cell cycle at S-phase, and reduces tumor formation in mice. Mechanistically, HIGD2A depletion decreases cellular ATP levels, impairs mitochondrial function, and attenuates MAPK/ERK pathway activation. Targeting HIGD2A may offer a new therapeutic strategy for HCC.
