Summary: Cholangiocarcinoma (CCA), a type of epithelial cancer, has poor outcomes and is often diagnosed late. The role of plasminogen activator, urokinase (PLAU), in CCA has not been well-studied. This study used data from Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) database, as well as immunohistochemical staining, to determine the expression of PLAU in CCA. PLAU was found to be significantly higher in CCA tissues compared to normal tissues, and high PLAU expression was associated with poor patient survival. Functional assays in vitro and in vivo showed that silencing PLAU suppressed CCA cell proliferation and migration while promoting apoptosis. Additionally, the NF-κB signaling pathway was found to be involved in PLAU-induced malignant phenotypes in CCA cells. Inhibiting high PLAU expression in CCA may be a potential targeted therapy approach.
