Micro-RNAs 518d-3p and 618 Are Upregulated in Individuals With Type 1 Diabetes With Multiple Microvascular Complications

Objective: To compare the serum micro-RNAs (miRNAs) profile of individuals with type 1 diabetes without microvascular complications vs. those with multiple severe microvascular complications, in order to identify epigenetically modulated pathways in these two groups of individuals. Research Design and Methods: A total of 10 subjects were selected among individuals followed in the Diabetes Outpatient Clinic and sorted according to the absence or presence of all microvascular complications. Samples from these participants were used for evaluation of serum miRNA expression profile employing a qRT-PCR assay with hydrolysis probes based on the Taqman Low Density Arrays (TLDA) system. The top six most differentially expressed miRNAs between the aforementioned groups were validated by qRT-PCR in additional 47 type 1 diabetes individuals sorted according to the absence or presence of all microvascular complications and matched for age, sex, degree of metabolic control, diabetes duration, and age at diagnosis. Results: Twenty one out of three hundred and seventy seven miRNAs were upregulated in the group of individuals with all microvascular complications vs. the group without complications. The following miRs were validated: 518-3p, 34a-5p, 126-5p, 425-5p, 618, and 139-5p and logistic regression analyses showed that miRNA-518-3p and miRNA-618 were positively associated with multiple microvascular complications after adjustment for age, sex, diabetes duration, HbA(1)c and use of statin, angiotensin-converting enzyme inhibitors and amlodipine. Conclusions: In this cohort of type 1 diabetes individuals, serum miR-518d-3p and miR-618 were upregulated in those with diabetes kidney disease, diabetes retinopathy, peripheral neuropathy, and cardiovascular autonomic neuropathy in comparison to individuals with no microvascular complications.

Summary: This study evaluated the intestinal transcriptome response to Roux-en-Y gastric bypass (RYGB) in obese women with type 2 diabetes (T2D) and found that changes in the intestinal transcriptome may be associated with T2D remission and glycemic control. The analysis of differentially expressed genes (DEGs) revealed significant correlations between specific genes and glycemic markers, particularly in the duodenum. These findings suggest a potential role of intracellular cholesterol and lipid homeostasis in glucose control after RYGB.

Summary: This study evaluated the relationship between mitochondrial DNA replication, oxidative stress, and gene expression related to diabetes. The results showed a positive correlation between mtDNA copy number and tissue autofluorescence, as well as a negative correlation between PINK and PRKN gene expressions with age. The expression of mitochondriogenesis genes and TXNIP was higher in individuals with type 2 diabetes mellitus.

Summary: This study performed miRNA profiling of patients before coronary artery bypass grafting (CABG) surgery to identify miRNAs that could predict Vasoplegic syndrome (VS). Six miRNAs were increased and two were decreased in patients who developed VS compared to those who did not. The combination of hsa-miR-30d-5p and hsa-miR-770-5p showed high accuracy in predicting VS.

Summary: This study explores the proteomics of HDL subfractions in type 1 diabetes (T1D) and its association with clinical variables and subclinical atherosclerosis. Different protein expressions were observed in HDL2 and HDL3 of T1D patients compared to controls, and these proteins were related to lipid metabolism, inflammation, and antioxidant response. HDL proteomics was found to be predictive of subclinical atherosclerosis in T1D.

Summary: Diabetes mellitus is a risk factor for dementia and can induce neuronal dysfunctions through inflammation and other processes. This study found that obese individuals with diabetes had reduced GLUT4 expression and neuronal soma area in the hippocampus. Inflammatory activation decreased GLUT4 expression and negatively affected genes related to neuronal function. Inhibiting HDAC3 could potentially improve GLUT4 expression and counteract the negative effects of inflammation.

Islet transplantation: overcoming the organ shortage

Summary: Background: Type 1 diabetes mellitus (T1D) is a condition caused by autoimmune destruction of pancreatic beta cells. Transplantation of isolated pancreatic islets from donors is a potential treatment for T1D, but the limited availability of suitable pancreas donors is a major challenge. Methods: A retrospective study was conducted to analyze the profile of brain-dead human pancreas donors and the reasons for organ refusal. Results: The main reasons for refusal were hyperglycemia, technical issues, age, positive serology, and hyperamylasemia. Conclusions: This study provides insights to improve the availability of suitable pancreas donors and enhance the outcome of islet isolation and transplantation.

Summary: The study examined the expression of inflammation-related miRs bound to HDLs in breast cancer patients and controls. It was found that HDL from breast cancer patients had higher anti-inflammatory activity compared to controls, especially in advanced stages of the disease. The miRs 223-3p and 375-3p in HDLs were significantly higher in breast cancer patients and showed high discriminatory power between breast cancer and controls.
