Summary: In this study, WNT5A protein was found to be significantly downregulated in BM tissues and EGFR-mutant samples of NSCLC patients, with overexpression inhibiting the growth and migration of EGFR-mutant cells. Further research revealed that WNT5A is negatively regulated by E2F1 and that its repression is dependent on the ERK1/2 pathway in EGFR-mutant cells. These findings suggest that targeting the ERK1/2-E2F1-WNT5A pathway could be an effective strategy for treating BM in EGFR-mutant NSCLC.

Summary: This study found that MGST1 is upregulated in gastric cancer and is associated with poor overall survival of patients. Knockdown of MGST1 significantly inhibits gastric cancer cell proliferation and cell cycle by regulating the AKT/GSK-3β/β-catenin axis. Additionally, MGST1 inhibits ferroptosis in gastric cancer cells.

Summary: This study found that the expression of IRF-2 was downregulated in gastric cancer tissues and was negatively correlated with the prognoses of gastric cancer patients. IRF-2 inhibits tumor growth by regulating the transcription of AMER-1 and the Wnt/β-catenin signaling pathway.

Summary: In this study, TRIM21 was identified as a tumor-suppressive gene in gastric cancer through analysis of the TCGA database. It was found to be involved in apoptosis, cell proliferation, and the JAK/STAT signaling pathway. TRIM21 expression was positively correlated with infiltration of various immune cells. Further experiments confirmed its inhibitory effects on proliferation and migration, as well as promotion of apoptosis in gastric cancer cell lines. The results also revealed that STAT1 inhibited the transcriptional activity of TRIM21.

Summary: The study investigated the impact of Wnt/beta-catenin inhibition on endometrial cancer recurrence, with PRI724 and SM04690 showing significant inhibitory effects on CTNNB1-mutant cell lines. Targeting the Wnt/beta-catenin pathway effectively inhibited proliferation and beta-catenin/TCF transcriptional activity in CTNNB1-mutant EC, suggesting a potential therapeutic vulnerability for treatment of CTNNB1-mutant EC.

Summary: The study explored the pathological role of a nonhistone methyltransferase in gastric cancer (GC), identified EEF1AKMT3 as a novel tumor suppressive methyltransferase, and revealed its mechanism of action through regulating the methylation of MAP2K7 and the degradation of TP53. The findings suggest potential targeted therapy in GC by manipulating the dysregulated EEF1AKMT3/MAP2K7/TP53 signaling axis.

Summary: This study investigated the role and mechanism of DVL in gastric cancer development, and found that DVL2 is highly expressed in gastric cancer tissues, which is significantly correlated with clinicopathological parameters and patient survival. Inhibiting DVL2 can suppress gastric cancer cell proliferation, migration, and invasion, suggesting that DVL2 may be a new therapeutic target for gastric cancer.
