CYTOTOXICITY AND GENOTOXICITY OF IRON OXIDE NANOPARTICLES: AN IN VITRO BIOSAFETY STUDY

With the development of nanotechnology and the wide use of iron oxide nanoparticles, it has become necessary to assess the potential adverse biological effects of magnetite. This study investigated the cytotoxicity, genotoxicity and oxidative damage of different concentrations of magnetite (0 to 1000 mg/L) in human whole blood cultures. After supplementation of magnetite, the blood samples were incubated for 72 h. Cell viability was assessed by the 3-(4,5-dimethylthiazol-2-yl) 2,5-diphenyltetrazolium bromide (MTT) and lactate dehydrogenase (LDH) release assays. The total antioxidant capacity (TAC) and total oxidant status (TOS) were determined to evaluate the dose-dependent effects of magnetite on the oxidant/antioxidant balance and to evaluate the potential oxidative injury due to increased oxidative stress. Genotoxicity was estimated by by the sister chromatid exchange (SCE), micronuclei (MN) and chromosome aberration (CA) assays and determination of 8-oxo-2-deoxyguanosine (8-OH-dG) levels. The results of MTT and LDH assays showed that the higher concentrations of magnetite (100, 150, 300, 500 and 1000 mg/L) decreased cell viability. Concentrations of magnetite higher than 10 mg/L increased TOS levels and decreased TAC levels in human blood cells. Increasing concentrations of magnetite caused significant increases in MN, SCE and CA rates and 8-OH-dG levels. The obtained results showed that magnetite exerted dose-dependent effects on oxidative damage, genotoxicity and cytotoxicity in human blood cells.

Multiple lipid nanoparticles as antimicrobial drug delivery systems

Summary: This study aimed to improve the antibiotic activity of vancomycin hydrochloride (VAN) by formulating multiple lipid nanoparticles (MLNs) containing carvacrol (CAR) and VAN co-loaded (CAR-VAN-MLNs). The results showed that the drug-loaded MLNs had appropriate particle size, negative surface charges, and high encapsulation efficiencies for both CAR and VAN. The stability and sustained drug release of the formulations were confirmed, and the morphological analysis indicated that the MLNs had a spherical shape. The co-encapsulation strategy improved the antibiotic activity of VAN against Staphylococcus aureus.

Summary: Pancreatic cancer has limited response to current therapeutic options, so there is a need for more effective and low-toxic agents. In this study, the antiproliferative activities of newly synthetized resveratrol analogues in pancreatic cancer cell lines were explored. The most active compound exhibited marked antiproliferative effects through mechanisms involving DNA damage, apoptosis induction, and interference in cell cycle progression.

Emerging Effects of Resveratrol Derivatives in Cells Involved in Oral Wound Healing: A Preliminary Study

Summary: Recently, research has focused on finding new approaches to manage oral wound healing. The use of resveratrol (RSV) as a drug is limited due to its poor bioavailability. This study aimed to investigate a series of RSV derivatives (1a-j) with better pharmacokinetic profiles. Among them, derivatives 1d and 1h showed increased cell viability compared to RSV. Further studies demonstrated that both 1d and 1h exhibited positive effects on cell viability, proliferation, and gene expression in various cells involved in oral wound healing. Additionally, 1d and 1h showed promising physicochemical properties and stability, indicating their potential as RSV-based agents for oral tissue repair.

However, physical barriers, hostile microenvironment, and tumor heterogeneity limit their efficacy against solid tumors. The use of CAR-T-derived EVs as therapeutic agents may enhance CAR-T homing and overcome some adverse effects.

Novel styryl-thiazole hybrids as potential anti-Alzheimer's agents

Summary: In this study, a series of novel styryl-thiazole hybrids were designed, synthesized, and evaluated as potential candidates for the treatment of Alzheimer's disease. Among the synthesized hybrids, compounds 6e and 6i showed promising results in increasing cell viabilities and inhibiting AChE. Docking studies and in vitro assays further confirmed these findings. Compound 6i exhibited better physicochemical properties and demonstrated potential as a multi-targeted active molecular profile for treating AD.

The prognostic and predictive values of differential expression of exosomal receptor tyrosine kinases and associated with the PI3K/AKT/mTOR signaling in breast cancer patients undergoing neoadjuvant chemotherapy

Summary: The study aimed to compare the expression levels of receptor tyrosine kinases (RTKs) and AKT1 and mTOR in the exosomes of breast cancer (BC) patients before and after neoadjuvant chemotherapy (NACT) with healthy females. The results showed that FGFR2, PDGFRB, AKT1, and mTOR were significantly upregulated in BC patients before NACT compared to the healthy group, while PDGFRB and AKT1 were significantly down-regulated after NACT. PDGFRB expression level correlated with therapy response and tumor size.
