Utilization of Natural Detergent Potassium Laurate for Decellularization in Lung Bioengineering

Perfusion decellularization by detergents is used to harvest native organ scaffolds with delicate three-dimensional structures from the natural extracellular matrix (ECM). However, detergents often damage ECM proteins, disrupting the reseeding of cells during the bioengineering process. In this study, we analyzed the ability of the natural soap potassium laurate (PL), generated from a natural fatty acid, to act as a detergent for lung bioengineering. Harvested rat lungs were decellularized by PL or the conventional detergent sodium dodecyl sulfate (SDS). The results indicated that PL could remove cells and DNA. Moreover, PL-decellularized lungs showed obviously improved preservation of architecture, with elastin microfibrils, sulfated glycosaminoglycans, and ECM proteins, compared with that with SDS. Mesenteric implanted PL-decellularized scaffolds demonstrated significantly reduced inflammatory reactions compared with SDS-decellularized scaffolds. In vitro, rat lung microvascular epithelial cell (RLMVEC) proliferation and survival rates were increased by coculturing with minced PL-decellularized scaffolds but not minced SDS-decellularized scaffolds. After recellularization using RLMVECs and adipogenic stem/stromal cells, recellularized cells uniformly distributed in PL-decellularized scaffolds but not in SDS-decellularized scaffolds. After the transplantation of bioengineered lungs, lung congestion was obviously reduced with PL-decellularized bioengineered tissues. Overall, our findings suggest that PL is very well suited for organ bioengineering. Impact Statement Recent advances in tissue engineering using decellularized organ scaffolds have expanded the possibilities for organ replacement therapy. However, detergent-based decellularization itself damages the extracellular matrix (ECM), which results in failure associated with the transplanted bioengineered organ. This study determined that potassium laurate (PL), a natural detergent, significantly reduces lung ECM damage during the decellularization process compared with protocols using sodium dodecyl sulfate. PL-decellularized lungs showed better microarchitecture preservation and low biological reactions after subcutaneous implantation. PL-decellularized scaffolds supported rat lung endothelial cell attachment/proliferation and the bioengineered lungs significantly reduced lung congestion after transplantation.

Cadaver surgical training in Japan: its past, present, and ideal future perspectives

Summary: The framework for cadaver surgical training (CST) in Japan was established in 2012 and is currently practiced in 33 medical schools and universities. The CST Promotion Committee of the JSS reviews and provides guidance for CST reports submitted by schools. In the future, an operating organization funded by stakeholders will be needed to sustain and oversee CST implementation.

Beneficial Impact of Interspecies Chimeric Renal Organoids Against a Xenogeneic Immune Response

Summary: We generated interspecies chimeric renal organoids by adding heterologous rat renal progenitor cells to single cells from mouse fetal kidneys and observed their differentiation into renal tissues of three lineages. Chimeric renal organoids with a higher proportion of recipient cells showed reduced rejection compared to complete xenograft organoids.

Summary: This study compared the transplant rejection between neonatal and fetal pig kidneys in renal heterotransplantation. The results showed that fetal pig kidneys were spared from rejection despite the administration of the same immunosuppressive protocol to the monkeys and the recipient blood vessels flowing into the fetal kidneys.

Summary: Lung cancer is the leading cause of cancer-related death worldwide and accounts for about 40-50% of brain metastases. In early-stage lung cancer, the incidence of brain metastases is around 3%. The current international guidelines have different recommendations regarding brain imaging for early-stage cancer patients. Some studies suggest that surgical or radiosurgical treatment may improve survival, especially in asymptomatic patients. Advances in genome analysis have identified molecular targets for pharmaceutical agents. This multicentric study analyzed data from 577 patients with early-stage lung cancer who underwent brain imaging.

Assessment of reports on cadaver surgical training (CST) implementation in Japan: current status and challenges

Summary: This article reports on the contents and conflicts of interest of Cadaver Surgical Training (CST) programs in Japan based on professional autonomy. The Japan Surgical Society CST Promotion Committee evaluated the validity of the reports from participating universities and provided advice on proper implementation of CST. Over the past decade, 38 universities have been using cadavers for clinical education and research, such as CST. In case of identified inappropriate entries or unclear areas, the universities could be asked to resubmit their reports. This review clarifies the evaluation points of the CST reporting system, presents the status and problems of its reporting, and provides suggestions for future dissemination of CST.

Summary: The number of patients with end-stage renal failure is increasing annually worldwide, but there is a shortage of renal transplantation donors. Transplanting renal progenitor cells into heterologous fetuses has been shown to induce the development of nephrons. In this study, researchers successfully generated human-mouse chimeric kidneys by combining this technology with drug-induced removal of specific renal progenitor cells in transgenic mice. They also developed a model using re-aggregation of mouse renal progenitor cells to evaluate the chimerogenic potential of human nephron progenitor cells derived from pluripotent stem cells. This study provides promising potential for the evaluation and generation of chimeric kidneys for regenerative medicine.

Preliminary Observations of An Ex Vivo Normothermic Whole Blood Machine Perfusion in An Experimental Liver Transplant Porcine Model

Summary: This study explored the feasibility of using livers from uncontrolled donors after circulatory death (u-DCD) through transplantation. A pig model simulating u-DCD was used, and liver function was evaluated after 24 hours of perfusion. The results showed that u-DCD livers can be successfully transplanted using ex vivo machine perfusion.

Pre-Administration of Blackberry Extracts in Induced Ischemia Reperfusion Events in Rodents
